英文摘要 | Adult Culicoides arakawae were collected from chicken farm by New Jersey light trap. Bloodsucking and gravid females were separated in laboratory, and then reared in a 25 ℃ incubator for 3 days. The mean survival rate(64.2%) of midges in 80% constant humidity was significantly higher (p< 0.001) than (32.3%) reared in fluctuatina humidity. Gravid females induced to lay eggs on damp filter paper by confining individual insect in small vials with a piece of wet paper inside for oviposition showed 49.6% mean oviposition rate, and 57.3 mean egg number. However, those individually confined in petri dish containing 1.2% agar showed 72.3% mean oviposition rate, and 79.4 mean egg number. The latter not only provide higher efficiency to induce oviposition than the former (p<0.05), but could also be easily carried out. Adult midges, colonized in laboratory, were also reared in 25 ℃ incubator. Their development over 12 days duration compared with those under 12 days, the emergence rate of pupa, survival rate of adults and blood feeding rate of females all significantly declined (p<0.05). However, the adults with their larval period of over 12 days, reared at 80% constant humidity or in the cup with three bottom layers, showed significantly improved survival and blood feeding condition (p<0.05). The survival and blood feeding of adults reared in the cup with three bottom layers, were significantly higher than those adults reared at 80% uniform humidity (p<0.05). The former showed the highest survival rate and blood feeding rate, 78.7% and 77.9%, respectively. Our results showed that properly controlled humidity could improve the rearing efficiency of larvae and first generation adults of C. arakawae. Efficient breeding and blood feeding of C. arakawae, the vector of leucocytozoonosis, could be beneficial for studying the relationship between the vector and protozoan. |
