中文摘要 | 第2型豬鏈球菌(Streptococcus suis type 2)是一種最常由豬腦膜炎、關節炎及多發性漿膜炎分離出來的細菌;也會引起人的腦膜炎,屬重要的人畜共通傳染病。由於豬鏈球菌各血清型(35種莢膜抗原型)中,以第2型普遍的存於健康帶菌或發病豬群;因此,常無法進行流行病學分析,去追查感染源。若能有效分辨豬鏈球菌種(species)以下之型別,期望有助於流行病學上之研究。本實驗利用脈衝電場電泳限制�t分析(pulsed-field gel electrophoresis, PFGE)、隨機增殖聚合�t鏈反應(randomamplification of polymorphic DNA, RAPD)以及核醣體核醣核酸基因型別法(ribosomal DNA RFLP analysis, Ribotyping)分析比較32株野外分離株(Type 2)及1標準菌株。應用PFGE且選用Smal及Sfil酵素做切割時,均各別獲得6種不同圖譜;以引子(primer)CAM 2及CAM 2+CAM 3配合RAPD可分別得到8及6種不同圖譜;而以Bam Hl核醣體核醣核酸基因型別法進行分析則只獲得4種不同圖譜。同為腦來源之菌株在Sfil酵素切割之PFGE圖譜具很高之相似性,肺來源者於Smal及Sfil酵素切割之PFGE圖譜也分別具相似性。分離株之來源與隨機增殖聚合�t鏈反應及核醣體核醣核酸基因型別法分析結果則無明顯關聯。此顯示脈電場電泳限制�t分析為豬鏈球菌流行病學研究上極具潛力之方法。 |
英文摘要 | Streptococcus suis type 2 is frequently isolated from young pigs suffering from meningitis, arthritis and polyserositis. It is also known to cause meningitis in humans; thus, it is an important zoonotic disease. Among 35 capsular types, the type 2 strains are widespread both in clinically healthy and diseased pigs; therefore, it is difficult to correlate between strains origin and the disease by epidemiological studies. To make subspecies differentiation or even strains discrimination under the same capsular type is therefore a matter of great concern. A collection of 33 Streptococcus suis type 2 strains, 32 by field isolates and 1 reference strain, were examined by pulsed-field gel electrophoresis (PFGE), random amplification of polymorphic DNA (RAPD) and ribosomal DNA restriction fragment length ploymorphism analysis (Ribotyping). The PFGE restriction analysis with both Smal and Sfil generated 6 patterns among the 33 strains. RAPD with primer CAM2 and CAM2+CAM3 resulted in 8 and 6 distinct patterns, respectively, 4 patterns were obtained with BamH I Ribotyping. Most of the Sfil PFGE restriction analysis profiles of the brain isolates were similar to one another, moreover, most of the Sfil and Smal PFGE restriction analysis profiles of the lung isolates were also similar. Apparently, PFGE combined with restrction enzyme analysis provides a more discriminatory method for subspecies differentiation of these organisms. |
