中文摘要 | 家禽流行性感冒(Avian influenza, Al)與新城雞病(Newcastle disease, ND)在臨床症狀上有許多類似之處,為了建立快速檢測並區分上述兩種疾病之技術,本實驗首先從A型流行性感冒病毒(Avian influenza, AIV)核蛋白基因具高保留性片段之序列設計一對引子,用已知亞型(H1至H13亞型)之AIV標準株,以及野外分離之AIV共77株, 以反轉錄聚合�t連鎖反應(RT-PCR)進行測試,結果證實此對引子可從上述所有的AIV病毒株中增幅出一段330bp片段,並具高度的特異性與敏感性。其次,我們將此對引子配合先前已開發成功能偵測新城雞病病毒(Newcastle diseasevirus, NDV)之引子,進行單管多引子 RT-PCR(Multiplex RT-PCR),則可檢測並區分AIV與NDV,並且還能區分NDV之疫苗株與野外株。此單管多引子RT-PCR技術可簡化檢測AIV與NDV時之工作量及檢驗成本,很適合作為快速診斷之用。 |
英文摘要 | Avian influenza and Newcastle disease share many common clinical symptoms. A method for rapid identification and differentiation of these two diseases comes to be necessary. A pair of primers was designed based on the nucleotide sequences of the highly conserved region of nucleoprotein of avian influenza virus. By reverse transcription-polymerase chain reaction (RT-PCR), this pair of primers could amply a 330 bp fragment from 13 reference strains (H1-H13) and 77 field isolates of avian influenza virus with a high sensitivity and specificity. A multiplex RT-PCR procedure was then developed by using the above AIV primers and primers for the detection of Newcastle disease virus (NDV) developed in the previous study. This multiplex RT-PCR procedure could detect both AIV and NDV in a single reaction tube, and could differentiate vaccine strains from field isolates of NDV. Since the one tube multiplex RT-PCR could significantly reduce the time and cost of AIV and NDV, it is suitable for rapid identification and differentiation of AIV and NDV. |
