中文摘要 | 我們以往的研究證實,30%紅斑狼瘡患者血清內可出現干擾素,而正常人僅4.5%。本研究擬以狼瘡病患者之淋巴球利用PHA,Con A及中藥來刺激,瞭解其干擾素產生能力,並與正常人相比較。中藥在動物試驗中已證實有干擾素誘發能力之四種中藥分別為當歸、黃耆、冬瓜子以及仙鶴草。11例經臨床及實驗室診斷證實為狼瘡病患者及15例正常健康者進行上述研究。結果顯示:以Con A刺激,8例狼瘡病患者有4例其淋巴球無法分泌干擾素,而正常5例全部都可刺激分泌;以PHA當作刺激物,效果與Con A類似,但干擾素分泌量却不如Con A多;如利用上述4種中藥作刺激物,無論患者及正常者組沒有一種可使淋巴球分泌干擾素。我們結果顯示狼瘡病患者之淋巴球有免疫缺陷,以致用PHA及Con A刺激干擾素分泌能力均比正常者差。另外,中藥是否在體外可作為很好之干擾素誘發物,尚需更進一步之實驗證實。 |
英文摘要 | Our previous study demonstrated interferon was present in 30% of systemic lnpus erythematosus (SLE) patients' sera, which was significantly higher when compared to 4.5% in the normal control. In the present study, peripheral blood lymphocytes were isolated from SLE patients and stimulated with T cells mitogen (PHA, Con A) as well as herbs to see its interferon production. Herbs which we previously tested to have an interferon inducing response were 1. Angelical sinesis Diels. 2. Astraglus membranaceus Bage. 3. Benincasa hispida Cogn. 4. Agrimonia pilosa Lederb. Within one year, 11 cases with definitive diagnosis of SLE were enrolled in this study. Fifteen normal healthy adults were used as a control. The results showed interferon from Con A stimulated lymphocytes was only detectable in 4 of 8 lupus patients, whereas in the control group, interferon appeared in all 5 cases. The interferon titer was significantly higher in the control group than in lupus patients. PHA had a similar effect as Con A to induce interferon production, but in general, a lower interferon titer was noticed as compared to Con A. The above 4 Chinese herbs have been tested but unfortunately, none could induce lymphocyte interferon production either in lupus patients or healthy individuals. Our findings suggest the deficient interferon production from SLE mononuclear cells by Con A or PHA stimulation may be due to immunodeficiency of lymphocytes. Whether herbs in vitro could serve as interferon inducers still needs to be further examined. |
