英文摘要 | Differential display of mRNA is a recently described technique using reverse transcribed polymerase chain reaction (RT-PCR) to compare cDNA from paired healthy and malignant tissue to determine whether there is either genetic loss (putative tumor suppressor gene) or overexpression (putative oncogene) in the malignant cells. Using the differential display technique for non-small cell lung cancer (NSCLC), we isolated 10 candidate bands, and seven bands were sequenced. One of the seven bands, 244 bp, had an identity of 100% to part of a cloned human DNA sequence, which map to chromosome 11p13. Five (56%) of nine patients with NSCLC revealed overexpression by using the same primer set for RT-PCR. Another 102 bp band was preferentially overexpressed in healthy lung tissue, but not in non-small cell lung cancer tissue. The band was identical to known mRNA sequences of a human homeobox gene. We suggest that differential display is a rapid and sensitive method for identifying genes that are differentially expressed in NSCLC. |
