中文摘要 | 本研究利用逢機複製DNA片段多態性(Random amplified polymorphic DNA, RAPD) 分析方法,針對臺灣省畜產試驗所近親品系土雞之遺傳組成進行分析,以便提供上雞選育之 遺傳標記。 試驗中採取 4 個近親品系 L7、9,II 及 12 之血液,以 DNA 萃取套組萃取全 血之基因組 DNA, 測定並調整適當濃度後,再以合成之 10 鹽基對 (10 bp) 寡核��酸作為 引子, 以不同土雞個體之 DNA 作為模板進行聚合醃連鎖反應 (po1ymerase chain reaction, PcR)。PCR 複製後之產物以 1.5% 瓊瓊脂糖膠體進行電泳分析。本試驗利用 250 種不同寡核��酸序列之逢機引子分析土雞之 DNA,並將可以偵測不同個體及品系間 RAPD 指 印遺傳多態性之結果,在掃描影像後利用群組分析電腦軟體計算品系間及品系內之相似性。 試驗結果顯示,以 RAPD 方法分析近親土雞之遺傳組成,可以利用 RAPD 指印之相似性來估 算品系內及品系間之遺傳之相關性及變異性。 L7 與其它三個品系間之相似性較低,其遺傳 距離他最遠。四個土雞近親品系與來亨雞間之遺傳距離則更遠。 |
英文摘要 | The purpose of this study was to analyze the genetic similarity of highly inbred lines of Taiwan native chicken in Taiwan Livestock Research Institute by random amplified polymorphic DNA (RAPD) method. Four hundred and eighty blood samples were collected from each of inbred lines (line 7,9, II, 12) and white Leghorn. Genomic DNA was extracted and detected for the concentration as well as used as template. Two hundred fifty single short (10 bp) oligonucleotide were used for RAPD analysis. The PCR reaction products were separated by electrophoresis in 1.5% agarose gels and stained with ethidium bromide to visualize amplified products. The polymorphic results were scanned and analyzed by Gelcompar software. The genetic correlation and variation within and between inbred chicken lines were estimated. The results showed that RAPD analysis provided a rapid and effective method to detect the genetic variation of lines and individuals of chicken. Line 7 had lower similarity and farther genetic distance than other inbred lines. The genetic distance between inbred Taiwan native ckicken lines and Leghorn chicken was farther. |
