中文摘要 | 本研究利用逢機複製DNA片段多態性分析方法(random amplified polymorphic DNA, RAPD)分析不同家禽個體間的遺傳組成變異性。試驗中分別採取菜鴨、正番鴨、土雞及白色中國鵝之全血,以快速DNA萃取套組萃取紅血球中之基因組DNA,測定並調整濃度後,再以合成之10鹽基對(10bp)寡核?酸作為引子,以不同家禽個體之DNA作為模板進行聚合?連鎖反應(PCR)。PCR複製後之產物以1.5%瓊脂糖膠體進行電泳分析。本試驗利用40種不同寡核?酸序列之逢機引子分析褐色菜鴨、正番鴨、土雞及中國鵝之DNA。試驗結果顯示,以RAPD方法分析四種家禽之遺傳組成,可以有效區分品種、品系或個體遺傳組成的變異性,在能產生DNA複製片段的引子中,依家禽品種的不同,約有70%至95%的引子可產生多態性RAPD指印。部分引子之複製產物出現不同性別之特異RAPD指印。由於RAPD分析方法可以省去限制?分切片段長度多態性(RFLP)分析時之DNA轉漬、探針雜交與DNA用量較少等優點,在探討非特定基因之情形下,可利用本法大量且迅速分析不同動物個體間之DNA多態性,進而供經濟性能基因定位之標記,實為一理想的分析方法。 |
英文摘要 | The purpose of the studies was to evaluate the application of random amplified polymorphic DNA (RAPD) method to detect genetic variation in poultry. The poultry studied included Brown Tsaiya duck, Muscovy duck, Native chicken and Chinese geese. Genomic DNA was extracted and its concentration was detected, and used as template DNA. RAPD was detected using forty kinds of a single, short (10 bp) oligonucleotide. The PCR reaction products were separated by electrophoresis in 1.5% agarose gels and stained with ethidium bromide to visualize amplified products. The results showed that RAPD analysis provided a rapid and effective method to detect the genetic variation of species, lines and individuals of poultry. Among the primers tested, 70% to 95% of those which had DNA products could produce polymorphism in different species. Some female-specific RAPD products were also obtained in Different kinds of poultry with different primers. The main advantages of RAPD analysis over RFLP were: small amounts of DNA required, easily performed, rapidly to obtain the analysis result, avoiding problems of radioactive materials of Southern blot, probe hybridization, highly labor-intensiveness and costliness of RFLP analysis. RAPD method was suitable for the examination of large numbers of animal samples to investigate the non-specific locus and could be used as a genetic marker to facilitate the study of locus that are related to economically important traits. |
