中文摘要 | 兩個試驗選用52頭離乳仔豬,以探討飼糧中添加鉻對仔豬週邊血液單核細胞 (peripheral blood mononuclear cell,PBMC)分泌白血球中介質-2(interleukin-2,IL-2)活 性和注射脂多醣(lipopolysaccharide,LPS)對其生理反應之影響。試驗1於玉米-大豆粕-粉 頭基礎飼糧中添加□啶甲酸鉻的鉻0(對照組)或400ppb,每處理6隻3重複,由4週齡採任 食方式飼養至10週齡,測定PBMC於體外經伴刀豆球蛋白(concanavalin A,con A)和氯化鉻 刺激之IL-2活性。結果發現飼糧中添加鉻具有增加8週齡PBMC經con A刺激產生IL-2活性 的趨勢(P<0.10),且8和10週齡仔豬PBMC經con A刺激之IL-2活性皆以培養24h較12h為 高;而且,8週齡仔豬PBMC在12~24h培養期間之IL-2活性增量,以飼糧中添加鉻較對照組 者為高,惟後者PBMC培養基經添加con A或氯化鉻則反降。試驗2之鉻添加量和飼糧處理與 試驗1相同,每處理4隻2重複,由2週齡教槽開始飼養,於10週齡時靜脈注射LPS 1μg/kg BW後,測定其生理反應。結果發現添加鉻可降低血漿三酸甘油酯濃度;此外,豬隻注射LPS 後雖可增加血漿中腫瘤壞死因子-α(tumornecrosis factor-α,TNF-α)、皮質醇、葡萄糖及 尿素氮濃度,惟未受飼糧添加鉻之影響。綜合上述結果,離乳仔豬飼糧中添加400 ppb鉻之 □啶甲酸鉻可降低仔豬血漿中三酸甘油酯濃度,但不影響仔豬於LPS注射後之生理反應。 |
英文摘要 | Fifty-two crossbred weanling pigs were used to evaluate the effects of chromium (Cr) on interleukin-2 (IL-2) activity of peripheral blood mononuclear cells (PBMC) and the physiological response in weanling pigs injected with lipopolysaccharide (LPS) in two separate experiments. In Exp. 1, each treatment had a triplicate with six pigs for each replicate and the piglets were fed on experimental diets from 4 to 10 wks of age. The basal corn-soybean meal-wheat shorts diet was used which was supplemented with 0 or 400 ppb of Cr from chromium picolinate. The IL-2 activity of PBMC incubated with concanavalin A (con A) and CrCl3 were then measured on the pigs obtained at 8 and 10 wks of ages. The IL-2 activity of PBMC with con A induction tended to increase in group with Cr supplementation at 8 wks of age (P<0.10). The IL-2 activity of PBMC obtained from 24 hrs incubation was higher than those from 12 hrs incubation for con A treated groups both at 8 and 10 wks of age. At 8 wks of age, the difference of IL-2 activity between two incubation times was small for the Cr supplemented pigs. In Exp. 2, the Cr supplementation and basal diet were the same as in the Exp. 1. Each treatment was duplicated with four pigs per replicate and the experimental diets were given from 2 to 10 wks of age. Plasma metabolites, tumor necrosis factor-α(TNF-α), and cortisol were then measured on all pigs after i.v. injection of LPS 1 μg/kg body weight at 10 wks of age. Supplemental Cr decreased the level of plasma triglycerides. The levels of plasma TNF-α, cortisol, glucose, and urea nitrogen were not influenced by Cr supplementation, however, the levels were all enhanced after LPS injection. The result suggested that supplementation of 400 ppb Cr from chromium picolinate decreased the level of plasma triglycerides. However, Cr supplementation did not influence the physiological response in weanling pigs subjected to LPS injection. |
