中文摘要 | 高血壓疾病可能是造成血小板功能及血管反應改變的一個主因。本研究即是比較自發性高血壓鼠(SHR)及正常血壓鼠(WKY),在血小板凝集功能,血脂及離體胸主動脈,腸系膜動脈血管反應性之變化情形。 結果顯示SHR鼠的ADP所誘導之凝集反應,血中thromboxane B₂(TXB₂), 6-Keto-Prostaglandin Fl α (6-keto-PG F1α)濃度,TXB₂/6-Keto-PGF1α比值,及三酸甘油脂(triglyceride)含量皆比WKY鼠高,但高密度脂蛋白-膽固醇(HDL-C)較低。 SHR鼠胸主動脈對Norepinephrine (NE)及高鉀溶液的收縮反應皆小於WKY鼠。去內膜的腸系膜動脈對NE及高鉀的收縮反應,WKY鼠大於SHR鼠。完整或去內膜之胸主動脈對acetylcholin的舒張反應SHR鼠皆較低,完整的腸系膜動脈舒張反應WKY鼠也較高。 綜合以上結果SHR鼠thromboxane A₂及Prostacyclin量不平衡,及高血脂等現象可能是造成血小板活化,動脈硬化等症狀的原因之一,而SHR鼠不同部位血管反應性變化,顯示可能主要是由於大血管平滑肌及小血管的內皮細胞損傷或變化所造成。 |
英文摘要 | Hypertension may alter platelet function as well as vascular response. Our study is to compare the platelet aggregation, plasma lipids and vascular response of isolated aorta and mesenteric artery between spontaneously hypertensive rats (SHR) & normotensive Wistar-Kyoto rats (WKY). Our results showed that the maximal platelet aggregation (by height) induced by ADP, the plasma TXB2, 6-Keto-PGF1α, TXB2/6-Keto-PGF1α and triglycerides (TG) in SHR are all higher, while the plasma high-density-lipoportein cholesterol (HDL-cholesterol) is lower than WKY rsts. The vascontriction of aorta to NE and high K+ solution is decreased in SHR, however, the vaso-dilatation to acetylcholine was also lower in SHR. In the denuded mesenteric artery, the vasoconstriction to NE was enhanced in WKY rats. The relaxation response of intact mesenteric artery to acetylcholine was higher in WKY than in SHR. We conclude that the increase in platelet aggregation may be due to an imbalance between the concentration of TXA2 and PGI2; and the high TG but low HDL-cholesterol may promote atherosclerosis in hypertensive rats. The different vaso-response may be due to the differential effect of hypertension on different sites of arteries i.e., the main influence of great arteries is on the smooth muscles while of small arteries is on the endothelium. |
