| 中文摘要 | 本實驗目的在探討甲狀腺功能亢進情況下,對藥物之反應性,以進一步瞭解血管內膜與平滑肌功能在甲狀腺功能亢進時,引起血壓改變所扮演的角色。本實驗以離體之胸主動脈及腸繫膜動脈作為研究模式。同時比較上述兩種血管在內膜完整與去除內膜的情況下,對各種收縮劑,如NE、All與高鉀等;及舒張劑,如ACh及NG等之反應情況。實驗結果顯示,胸主動脈及腸繫膜動脈在內膜完整情況下,對NE之收縮反應較去除內膜時為低。同時上述兩種血管在內膜完整的情況下,ACh(一種內膜依賴性之舒張劑)之舒張血管作用,在甲狀腺功能亢進組較正常對照組之反應為差。由以上之結果,更加說明血管內膜的功能在甲狀腺功能亢進的情況下,可能受到某種程度之損傷。初步結果顯示在甲狀腺功能亢進的情況下,血管內膜可能在製造及釋放內膜舒張因子(如:EDRF及PGl₂等)之功能受到抑制:抑或內皮細胞製造或釋放EDCF(如:endothelin)增加。相反地,對於已知之內膜非依賴性收縮劑(高鉀)及舒張劑(NG)之反應,在甲狀腺功能亢進的情況下,血管反應性均較正常對照組為小。此結果更說明甲狀腺功能亢進時,藥物反應性之改變除影響血管內膜外,血管之平滑肌功能也受到相當程度的影響。綜合以上結果,初步認為甲狀腺功能亢進時,血管阻力改變除了受平滑肌之影響外,血管內膜也扮演著相當重要的角色。 |
| 英文摘要 | The responsiveness of vascular beds to various vasocontrictors and vasodilators in rabbit of hyperthyroidism was investigated in isolated intact and denuded aortic and mesenteric arteries. The hyperthyroidism animals were induced by administered subcutaneously with T3 (0.2 mg/kg/day) for 7 to 10 days, meanwhile, the control group was administered equal volume of 0.9% NaCl. Results demonstrated that the contractile responses to NE in intact aortic and/or mesenteric arteries were lower than those in denuded preparations. Furthermore, the vasodilator responses to acetylcholine in intact aortic and mesenteric arteries is deteroiated in hyperthyroidism animals. In contrast, the vascular responses to endothelium-independent agents (i.e. high-K or nitroglycerin) in arteries of hyperthyroidism were lower than those in the control group. These results demonstrated that the function of vascular smooth muscle in hyperthy-roidism was also altered. In conclusion, in addition to the alteration of vascular smooth muscle function, the endothelium may play an important role in the modulation of vascular resistance in hyperthyroidism. |
