中文摘要 | 延腦吻端腹外側升壓區(rostral ventral lateral medulla, RVLM)除了是血管運動中樞外,亦參與痛覺調控,而興奮性氨基酸(excitatory amino acid, EAA)也與血壓、痛覺的中樞調控皆有關。我們探討了SHR鼠(spontaneously hypertensive rat)的RVLM中NMDA(N-methyl-D-aspartate)受體對血壓的作用。在此研究中,我們以電刺激、多管玻璃電極局部微量注射興奮性胺基酸NMDA及NMDA受體拮抗劑AP5(2-amino-5-phos-phonovalerate)於延腦吻端腹外側,以探討延腦吻端腹外側升壓區在SHR鼠及WKY鼠的升血壓反應性及NMDA受體在感壓反射及對維持基礎血壓的角色。結果發現,以各種頻率的電及NMDA化學刺激SHR鼠延腦吻端腹外側升壓區的升血壓反應皆大於WKY鼠。AP5可拮抗NMDA及低頻率電刺激的升血壓反應,而對高頻率電刺激則無拮抗作用。且局部注射AP5後,低頻率電刺激SHR鼠及WKY鼠的升血壓反應無差異,但高頻率電刺激SHR鼠的升血壓反應皆大於WKY鼠。此結果顯示,SHR鼠延腦吻端腹外側升壓區的神經元及軸突具有較大的升血壓反應性。在另一方面,局部注射AP5於兩側SHR鼠及WKY鼠延腦吻端腹外側升壓區,皆會部份拮抗箝夾兩側頸動脈的升血壓反射,但對其基礎血壓則無影響。此結果表示,延腦吻端腹外側升壓區的NMDA受體雖參與感壓反射,但並非是持續活動的,與基礎血壓無關。 |
英文摘要 | We and others previously reported that NMDA receptor is involved in the pressure regulation. In the present study, we locally applied NMDA and electrical stimuli to the rostral ventral lateral medulla (RVLM) in the SHR and WKY rats. We found that SHR had a greater pressor reactivity than their normotensive controls. Local injection (4 nmole) of AP5 to the RVLM, did not affect resting blood pressure, however, did antagonize the electrically (5-20 Hz) evoked hypertension in SHR and WKY rats at 5-20 Hz electrical stimulation. The high blood pressure evoked by the high frequency stimulation (60 Hz) was not affected by AP5. These suggested that the hypersensitivity of RVLM in SHR involved both NMDA and non-NMDA mechanisms. In conclusion, our results revealed that SHR had a higher sensitivity to excitatory amino acid agonists in RVLM than their normotensive controls. |
