英文摘要 | Nitrate reductase catalyzes the rate-limiting step in the nitrate assimilation of cotton. In vivo assay with endogenous nitrate-N only as substrate best approximates nitrate assimilation. The necessity of analyzing numerous samples from multiple replications of field grown cotton prompted the development of a simple colorimetric determination of 0.1M k-phosphate buffer at pH 7.5 with 5% propanol and 1% chloramphenicol. Cotton petioles were sampled early in the morning, incubated in the medium under air for 1 h at 27°C. After reaction stoppage, the sample is retained in the incubation medium for 2 h. Ninety-eight percent of the nitrate-nitrogen (N)content in the petiole was found in the incubation medium, demonstrating that the method allows determination of the petiole nitrate-N content and its rate of redution. |
