中文摘要 | 以香葉醇與乙酸異戊酯於有機溶劑中進行酵素催化轉酯化反應為模式進行探討, 產物 - 乙酸香葉酯為香料中非常重要的成份。 源自 Candida cylindracea 之脂解酵素於 pH5.2 之緩衝溶液中與界面活性劑 -Span85 製備查界面活性劑包覆脂解酵素 (SCL), 用以 催化轉酯化反應。在 35 ℃下,[ 香葉醇 ]/[ 乙酸異戊酯 ] 比為 2 及酵素 / 界面活性劑 比為 0.5 時,反應 24 小時,於異辛烷及正己烷中轉酯化率分別為 68.3% 及 61.6%。此結 果遠高於逆微胞及粉末狀脂解酵素系統。 SCL 製備簡便,且酵素穩定性高;同時 SCL 在有 機溶劑中之溶解度相當低,使其具有易於回收及重複使用之特點。 |
英文摘要 | Transesterification of geraniol and isoamyl acetate in organic solvents was investigated. The ester product, geranyl acetate, is an important component of flavor and fragrance compounds. The lipase from Candida cylindracea interacted with the surfactant, Span85 in a buffer of pH5.2 to form surfactant-coated-lipase (SCL) for the transesterification reaction. At the condition of [geraniol]/[isoamyl acetate]=2, lipase/surfactant=0.5, in isooctane (logP=4.51) and n-hexane (logP=3.5) at 35 ℃, the transesterification yields after 24 h were 68.3 and 61.6%, respectively, which were much higher than those obtained by using reversed micelles and powdered lipase. The SCL was easy to prepare, exhibited long term stability and possessed low solubility in organic solvents. It could be beneficial fo rthe recovery and repeatedly used. |
