中文摘要 | 本研究探討Bacillus cereus在無菌鋁箔包裝保久乳之製程中引起的腐敗原因。 由B. cereus污染之腐敗包裝外觀皆是平酸腐敗,腐敗保久乳內容物pH值由6.6降至6.1, 而且產生苦味;腐敗調昧乳的內容物pH值由6.6降至4.3,而且產生濃稠、沈澱和酸敗,分 離並鑑定腐敗菌為B. cereus。B. cereus在含10 ppm Mn2+之nutrient agar培養基中,35 ℃培養產生之孢子耐熱性:D110℃為0.8分,如果再添加100 ppm Ca+和2.2M蔗糖之nutrient agar 培養基中,400C培養產生之孢子耐熱性:D110℃為2分。接種B. cereus之孢子於還原乳,以135 ℃,4秒和140℃,4秒之UHT殺菌,充填、包裝和保溫後,產品皆沒有腐敗產生;但若接種孢 子之還原乳於無菌管路中循環1小時,然後以CIP清洗,再以蒸汽殺菌管路後,仍然能在熱 交換板片及無菌桶前閥組上檢出殘留B. cereus,可見B. cereus之孢子能在CIP清洗和蒸 汽預殺菌後殘留在無菌系統內。 |
英文摘要 | The objective of this study was to investigate the spoilage caused by Bacillus cereus of aseptic packaged long life milk manufactured in aseptic processing and packaging plants. There is generally no difference in appearance between normal and spoiled packages of long life milk. The spoiled long life milk could have bitter taste, with pH value decreasing from 6.6 to 6.1, while the spoiled flavor long life milk becomes viscous with precipitation and sour taste, and the ph value decreases from 6.6 to 4.3. The spoilage microorganisms were isolated and identified by API 50CHB and API 20E to be Bacillus cereus. A sporulation condition for isolated Bacillus cereus under culture with nutrient agar containing 10 ppm MnCl2 at 35℃ resulted in the formation of spores with a D110℃ value of 0.8 min, while a sporulation condition with further addition of 2.2 M sucrose and 100 ppm CaCl2 at 40℃ resulted in an increase of the D110℃ value of 2 mm. The spores of Bacillus cereus were inoculated into reconstituted milk and their survival during UHT treatment was checked. It was found that they could not survive at 140℃ for 4 sec. But when the reconstituted milk inoculated with spores was circulated through plate heat exchanger and aseptic pipeline for 1 hr, and then subjected to CIP and pre-steam sterilization, we could find the survival of the spores on plate heat exchanger and the valve of aseptic tank. Thus, Bacillus cereus spores could survive processes such as disinfection and pre-steam sterilization in the pipeline. |
