中文摘要 | 弧菌感染不僅為水生動物常見的疾病之一,亦是食品公共衛生土重要病原菌,因 此弧菌的分離與快速鑑定就顯得格外重要。 自 200 組隨機引子進行多型性增幅聚合酵素連 鎖反應 (random amplified po1ymorphic DNA PCR;RAPD PCR), 嘗試建立各不同弧菌 chromosome DNA 之增幅多型性片段, 實驗結果顯示其中 11 組隨機引子可明顯區分包括 V.anguillarum、V. parahaemolyticus、V. vulnificus、 V. cholerae 與 V. damsela 等 幾種不同弧菌,此結果將可應用於各弧菌菌種間快速鑑別與臨床診斷之依據。另一方面,從 79 株弧菌分離菌株進行質體之分析, 發現具有質體之 V. anguillarum 其質體較呈一致性 , 並包含特有的質體衍生攝鐵系統基因 (plasmidmediated iron uptake system genes), 而其他不同弧菌之質體彼此間則差異性較大,此一結果方可作為 V. anguillarum 與其他弧 菌間鑑別之參考。 |
英文摘要 | Vibrio spp. are important pathogenic bacteria of aquatic animals and also can cause massive disease including enteritis, diarrhea and toxemia in human. In this study, random amplified polymorphic DNA polymerase chain reaction (RAPD PCR) was applied for establishing the DNA fingerprints of virulent Vibrio spp. Among 200 random primers, 11 primers were successfully utilized to distinguish Vibrio spp. such as V. anguillarum, V. parahaemolyticus, V. vulnificus, V. cholerae, V. tubiashii and V. harveyi. Moreover, several large plasmids with similar molecular size could be isolated from 79 Vibrio isolates. Of which, only V. anguillarum exhibited similar plasmid profiles with unique iron uptake system gene but there were significant molecular variations between different Vibrio species. The homogeneity of the chromosome fingerprints and plasmid profiles suggested that RAPD PCR can provide an accurate and rapid identification method comparable to the traditional biochemical assay. |
