英文摘要 | Flower heads of different developing stages in two different genotypes of Leucaena leucocephala, K67 and K488, were cultivated for the assessment of anther browning rate. A MS medium containing 10% coconut milk was used as basal media in the experiment. Observation was concentrated on the influence of different combination of auxins, sucrose concentration and photo-period on the anther browning. The results showed that genotypes had no effect on the in vitro reaction. There was positive relationship between anther browning and flower size under 10 hours lighting, i.e. lowest browning rate occurred in flower heads of 0.73-0.83cm in diameter. Pre-chilling of 4℃ for more than 24 hrs decreased browning rate in all conditions. However, treatment of pre-chilling of more than 24 hours tended to increase browning. Concentration of sucrose and different auxins appeared no significant difference on anther browning rate. Photo-period also influenced on the culture: dark and 10-hour light had the same effect whereas 24-hour light was observed to increase browning rate. |
