中文摘要 | 為了探討norathyriol提高內皮細胞的障壁功能及預防血管活性物質對細胞間障壁功能之干擾,本實驗研究norathyriol對於血清緊素在老鼠心臟內皮細胞引起大分子蛋白的通透及微絲纖維的改變之作用。加入norathyriol的內皮細胞可預防血清緊素及cytochalasin B所誘發的大分子蛋白通透。Norathyriol可抑制血清緊素在內皮細胞細胞骨架所引起的肌動蛋白纖維分佈之改變。10□M norathyriol對血清緊素所引起的肌凝蛋白聚集現象亦有抑制作用。結論,norathyriol降低血清緊素誘發老鼠心臟內皮細胞的通透性,是由於穩定肌動蛋白微絲纖維及改變肌凝蛋白的重組,此結果顯示血清緊素引起內皮細胞的通透增加是和細胞骨架的組成有關。 |
英文摘要 | To investigate the role of norathyriol in enhancing the endothelial monolayer paracellular barrier function, and preventing disturbance of the paracellular barrier function by vasoactive agents, this study examined the effect of norathyriol on serotonin-mediated macromolecule transfer and microfilament change in cultured rat heart endothelial cells (RHEC) .Incubation of the endothelial monolayer with norathyriol antagonized serotonin-and cytochalasin B-induced macromolecular permeability. Norathyriol also opposed the effect of serotonin on the distribution of actin filaments in the endothelial cytoskeleton. Upon addition of 10□6 M norathyriol, serotonininduced changes in the myosin and distribution of myosin were comparable to that of serotoninmonolayers. In conclusion, norathyriol decreased the permeability of RHEC caused by serotonin, which was associated with the stabilization of F-actin microfilaments and changes in the myosin organization, suggesting that influences on cytoskeletal assembly may be involved in this process. |
