英文摘要 | Epidermal growth factor (EGF) is one of the mitogenic proteins that can mediate cell differentiation, proliferation and regeneration. The renal tubular epithelium produces a large amount of EGF in the normal kidney, but urinary EGF is decreased in acute renal failure. To investigate the effect of EGF on renal tubular damage following renal ischemia, an experiment was undertaken in Male Sprague-Dawley rats undergoing 30 minutes of bilateral renal artery and vein clamp ischemia. The rats received exogenously administered EGF (25 ug) subcutaneously or the same volume of saline at 30 minutes after reperfusion. Blood was collected from the tail vein for creatinine determination. The mean creatinine level (mg/dl) at 48 hours following ischemia was 1.6��0.3 in the treated group compared with 3.3��0.7(p<0.05) in the group treated with only the vehicle. At 72 hours following ischemia, the mean creatinine level in the EGF treated animals was 0.6��0.l vs. 2.2��0.7(p <0.05) in the vehicle treated rats. In addition, none of the rats treated with EGF died while 2 of the 10 vehicle treated animals expired. There was less necrosis in the outer medulla of the kidney in the EGF treated animals (p<0.05). Body weight loss was significantly greater in the vehicle treated rats than in the EGF treated animals at 96 hours. Our results suggest that EGF can protect against the development of ischemic acute renal failure. |
