中文摘要 | 一、研究緣起及目的 Honokiol 及 magnolo1 是中藥厚朴的有效成分,它們皆 有大白鼠心肌粒線體的抗脂質過氧化作用,同時它們在大白鼠冠狀動脈結紮有抗心律不整的 作用,此種作用機轉至今尚未了解。 然而 nitric oxide 及 prostaglandin 皆能產生心血 管擴張及抑制血小板凝集作用, 本實驗設計是藉 nitric oxide 及 prostaglandin 之拮抗 劑與 honokiol 及 magnolo1 相互作用來探討 honokiol 及 magnolo1 在心肌缺氧時抗心律 不整的機轉。二、研究方法及過程 SD 大白鼠在 urethane 麻醉下,經由開胸術分離左冠狀 動脈, 在冠狀動脈結紮前 15 分鐘靜脈給予 10-7 g/Kg 的 honokiol 及 magnolo1 作為控 制組, 然而在 honokiol 及 magnolo1 治療前 15 分鐘分別給予 1OO mg/Kg acetylsalicylic acid 或 l mg/Kg 的 L- NAME 作為實驗組分別測定心律不整的現象。 三 、 研究結果及結論使用 honokiol 及 magnolo1 10-7 g/Kg 的劑量治療下, 如果先行給子 aspirin 則在 30 分鐘的缺氧期及 10 分鐘的血流再灌注期,其心律不整 (心室跳動過快及 顫抖 ) 皆有明顯的減少; 但如果先行給予 L-NAME 則此種抗心律不整的作用則消失, 因此 實驗證明在大白鼠冠狀動脈結紮下 honokiol 及 magnolo1 的抗心律不整作用也可能經由 nitric oxide 系統而非 prostaglandin 系統,其兩者雖皆能產生心血管擴張及抗血小板凝 集,但在生理上,前者是藉細胞內 cGMP 作用而後者是經由細胞 cAMP 的機轉。 |
英文摘要 | Honokiol and magnolol are active components of Magnolia officinalis. These were vasodilators and antiplatelet agents. these were found to reduce infract size and supress ventricular arrhythmias in rats subjected to coronary ligation. Nitric oxide and prostaglandin both derived from endothelium-derived relaxant factor(EDRF), are responsible for vasodilatation and inhibition of platelet aggregation. In this study, the mechanism of antiarrhythmic effects of honokiol and magnolol on underlying the differing pathway of EDRF in coronary ligated rats were investigated. Male rats were anesthetized with urethane. Acetylsalicyclic acid( 100 mg/kg) or No-nitro-L-arginine methyl ester(L-NAME, 1 mg/kg) was intravenously injected at 30 min before ligation of coronary artery. Honokiol or magnolol at dosages of 10-7 g/kg was intravenously administered 15 min before ligation of coronary artery as control. The incidence of ventricular arrhythmias induced by coronary occlusion was totally supressed with treatment of honokiol and magnolol. The durations of ventricular tachycardia and fibrillation were significantly reduced in the pretreatment with asprin during honokiol and magnolol administered compared to control. These antiarrhythmic effect of honokiol and magnolol during ischemia/ reperfusion period was abolished by L-NAME administered. We concluded that The antiarrhythmic effects of honokiol and mag- nolol may mediated through the different pathway of prostaglandin or EDRF/NO system. |
