中文摘要 | 子房柄培養為拯救種間雜交胚早期衰敗的一種可行辦法,本試驗以臺南選9號及 臺南 11 號兩栽培品種為材料,探討不同培殖體材料培養基對子房柄培養的影響。結果發現 含分生組織之子房柄為較佳的培殖體,授粉後 7 天比 10 天子房柄之生長表現為佳。MS 為 促進子房膨大的有效基礎培養基,添加 0.5mg/l KN 及 0.25 mg/l IAA 使臺南 9 號獲得最 高的發根及子房膨大率。高濃度 NAA 及 1mg/l BAP 可增進臺南 11 號的子房發育。然而, 大多數的培養基仍然無法產生較大的莢果及分化胚,因此如何經一次體外培養獲得成熟種子 或植株,仍然值得進一步的研究。 |
英文摘要 | In vitro culture of peg tips in Arachis is a feasible technique for recuing interspecific hybrids which often abort at early growth stages. The objective of this research was to evatuate effect of explants and media on in vitro culture of peg tips of A. hypogaea cv. TNS9 and TN11. The results shew that peg tips containing meristem were the favorable explant for in vitro culture. Seven-day-old peg tips had more growth responses than 14-day-old tips. The results indicated that MS medium was the favorable basal medium for ovary enlargement. Medium containing 0.5mg/l kinetin and 0.25 mg/l IAA had the highest frequency of rooting and ovary enlargement in cv. TNS9. High concentration of NAA induced large amounts of callus and suppressed peg elongation, root production and ovary development. NAA at 2 mg/l in combination with 1 mg/l BAP caused development of more ovaries in TN11. However, most of media did not produced larger pod and differentiated embryo. This suggests that further research will be required to obtain mature seeds or plants in a one-step in vitro cultrue. |
