| 中文摘要 | 大部分的絕對厭氣性真菌存在於反芻動物的消化道中,根據文獻顯示絕對厭氣性真菌對一般細菌與好氣性真菌無法利用的結晶性纖維素具有強的分解作用,對於反芻動物分解植物纖維素提供重要的幫助。本研究中分析測定六株分離自臺灣水牛、黃牛的絕對厭氣性真菌纖維素分解能力,並與Trichoderma viride與Asperigillus niger純化的纖維素分解酵素,進行纖維素分解能力的比較,當以濾紙作為整體纖維素分解能力測定基質時,由絕對厭氣性真菌中以Neocallimastix frontalis X-26菌株第六天培養上清液所測得的活性最高為0.020 U/mg protein,而經純化的T. viride與A. niger之纖維素分解酵素比活性分別為 0.035與0.015 U/mg protein;Endoglucanase的活性測定結果,絕對厭氣性真菌中以N. frontalis D-21菌株第六天培養上清液所測得的活性最高為0.460 U/mgprotein,而經純化的T. viride與A. niger之纖維素分解酵素比活性分別為0.545與0.148 U/mg protein。由此結果顯示絕對厭氣性真菌的培養液中具有很高的纖維素分解酵素活性,未來應用於纖維素分解酵素的生產極具開發潛力。 |
| 英文摘要 | Obligate anaerobic fungi are found in the gastrointestinal tract of the ruminants. The fungi have been known to possess diverse plant polysaccharide hydrolase activities. In this study, we develop a new approach to compare the cellulolytic ability with the crude supernatant of six Neocallimastix strains isolated from Taiwan native animals and two commercial cellulases purified from Trichoderma viride and Aspergillus niger. The cellulolytic activity of N. frontalis X-26 was 0.02 U/mg whereas the activity of T. viride and A. niger cellulase was 0.035 and 0.015 U/mg. Respectively, the endoglucanase activity of N. frontalis D-21 was 0.460 U/mg whereas the activity of T. viride and A. niger endoglucanase was 0.545 and 0.148 U/mg protein. These results suggest the obligate anaerobic fungi exhibit very high cellulolytic activity. |
