中文摘要 | 斑蝥素(Cantharidin)是從斑蝥(Mylabris phalerate Pallas)分離純化得到,其 在體外抗肝癌細胞株試驗中, 發現其具有明顯的抑制作用 (Hep 3B: IC �t =0.52 μ g/ml, HCC 36: IC �t =0.68 μ g/ml)。因此本實驗繼續針對其體內及體外之毒性進行評估 。在體外試驗方面,本實驗取自健康人之牙齦組織 (gingival tissue),分離出正常之纖維 母細胞 (fibroblasts), 檢測 cantharidin 對正常細胞的毒性。 結果顯示:cantharidin 對十株來自不同健康人之牙齦組織, 初代培養的纖維母細胞平均 IC �t =3.0 μ g/ml,治 療指數約為五倍。 體內之急性毒性,則以 cantharidin 1.3 mg/kg (LD �t ) 餵服 ICR 鼷 鼠後, 48 小時內,在 10 分鐘、30 分鐘、1 小時、4 小時、12 小時、24 小時、48 小時 犧牲鼷鼠。 測其血液中的白血球、紅血球和血小板, 及血清內 GOT, GPT, uric acid, creatinine 的生化值及器官變化。 結果發現與未服 cantharidin 之正常組 ICR 鼷鼠,並 無有意義的變化。 |
英文摘要 | Cantharidin was a strong inhibitory agent to hepatoma cell, which was isolated from spanish fly (Mylabris phalerate Pallas) in previous report. IC �t of Hep 3B and HCC 36 cell lines were 0.53 μ g/ml, 0.71 μ g/ml respectively. Therefore, we continune to investigate the toxic effects of cantharidin by in vitro and in vivo assay. The in vitro toxicity was adapted cell cytotoxicity to the primary culture normal human fibroblasts. They were isolated from the gingival tissues of 10 volunteers. The average IC �t is 3 μ g/ml and the therapeutic index is 5. The in vivo toxicity was oral administrated the LD �t dose (1.3 mg/Kg) to ICR mice. The biochemical changes of serum and organ weight were detected by the autohumalyzer (Human 850). The results of the acute toxic effects show no significant difference from control group. |
