中文摘要 | 根管治療充填劑中屬於樹脂類之充填劑如AH26與AH plus 充填劑,過去許多研 究顯示AH26根管充填劑會釋出formaldehyde而造成細胞毒性與基因毒性。AH plus 為新改良之充填劑,其特性與AH26充填劑相似,差異在於它不會釋出formaldehyde。 本研究的目的乃欲探討比較AH26與AH plus根管充填劑對人類口腔上皮癌細胞株之 細胞毒性研究。利用活細胞會吸收代謝3-[4,5-dimethyl(thiazol-2-yl)-2,5,-diphenyl- tetrazolium bromide]形成formazan之結晶,而於吸光器測得吸收光之原理(MTT 分析),藉由測其細胞之吸光值大小,即可得知細胞數之多寡來比較兩種充填劑之細胞 毒性。結果顯示:二種根管充填劑對人類口腔上皮癌細胞株均具有細胞毒性,50%致死 劑量(LD50)顯示AH26根管充填劑的濃度較AH plus根管充填劑的濃度低,以此證 明對人類口腔上皮癌細胞株而言,AH26 根管充填劑毒性比AH plus 根管充填劑毒性 強。 |
英文摘要 | The AH 26 and AH plus root canal sealers are the resin type filling material. The AH 26 root canal sealer will release the formaldehyde after mixing, but not the AH plus root canal sealer. The purpose of this study was to investigate the cytotoxicity of AH 26 and AH plus root canal sealers. By using the MTT assay, the oral cancer cell line-OC �� cell were tested. The result of the MTT assay showed that both the AH 26 and AH plus root canal sealers have cytotoxicity on OC �� cell. The AH 26 root canal sealer has hihger toxicity than the AH plus root canal sealer. The LD50 of the AH 26 and AH plus root canal sealers were as follow:22.7 and 23.9 u g/ml. What components of the AH plus root canal sealer will cause the toxicity are needed further investigation. |
