中文摘要 | 由野外萎縮性鼻炎患豬分離的D型巴氏桿菌產毒株製備粗製毒素(PMT cd)與類毒 素 (PMT cd toxoid),以小白鼠、天竺鼠及豬進行免疫試驗, 並評估其保護效力。 將 PMT cd 經 1% formalin 不活化後加入佐劑製備之類毒素菌苗,經不同動物免疫後,顯示皆有免 疫保護效果。小白鼠經不同劑量類毒素菌苗免疫後,分別以同株或不同株之相同血清型,或 不同血清型巴氏桿菌製備之 PMTcd 攻毒,結果顯示皆有保護效力,但以對同型同株之保護 效力最佳,50 ug 之免疫劑量就可達 100% 之存活率。 天竺鼠免疫後經不同濃度之 PMT cd 攻毒亦皆可耐受而無皮內出血壞死灶,免疫之天竺鼠於攻毒前皆可測得中和抗體力價。豬隻 免疫試驗顯示,不同劑量免疫組皆有保護效力, 惟 500 ug 免疫組尚有 50% 呈輕、中度鼻 甲骨萎縮病變,而 1000 ug 以上則達完全保護。 所有免疫組血清中和抗體力價與陽性對照 組比較皆有顯著差異 (P<0.05)。 本試驗發現自然感染 AR 豬隻無法測得中和抗體, 顯示 PMT 是不良的免疫原,雖能引致 AR 病變,但其免疫反應難以產生中和抗體。此外,被動免 疫試驗顯示免疫血清具有中和毒素的抗體可與毒素作用而達保護效力。 |
英文摘要 | The bacterin vaccine contained toxoid of Pasteurella multocida type D toxin (PMT) prepared to evaluate the immunogenicity and protective efficacy against swine atrophic rhinitis (AR) performed in mice, guinea pig and swine. The results indicated that good protection abilities were noticed on all experimental animals after receiving the formalin inactivated of PMT toxoid vaccine. The more applicable protection were observed when mice immunized and challenged the same isolates and/or serotypes of prepared PMT toxoid; also no mortality was found if vaccine contained the minimum dose of 50 ug PMT toxoid. Moreover, neutralizing titer of antibody could be detected and no dermonecrotic changes were found during the period of immunization in the groups of guinea pigs. The swine also had good immune response and the neutralizing titer showed significant variation (P<0.05) when compared with the non-vaccinated groups. However, 50% (3/6)of immunized pigs received 500 ug PMT toxoid still reveal mild to moderate lesions, but complete protections without clinical signs of AR were noticed after increasing the concentration to 1000 ug. These results suggested the prepared toxoid vaccine has good efficacy and vaccinated swine can mount significant immunity in contrast no neutralizing antibodies can be detected in natural infection of atrophic rhinitis in affected swine. |
