| 中文摘要 | 61天株齡木瓜植株經切芽處理後,其葉塊培植體可分化出再生木瓜小植株,但未 經切芽處理則無法分化出植株。木瓜植株於切芽處理後第 3 ∼ 10 天中,以第 8 天之第 7 葉序之葉塊培植體之植株再生率為最佳,當培養於含 2.2mg/l kinetin 及 4.7mg/l NAA 之 MS 培養基中,其植株再生率約 5.5%,顯示切芽處理對於木瓜葉塊培養之植株再生具促進作 用。以切芽處理之木瓜植株之葉塊培植體比較一段式及二段式葉塊培養法之植株分化率,則 以含 2.2mg/l kinetin 及 4.7mg/l NAA 之 MS 培養基中培養二週後,再移植至不含植物生 長調節劑之 MS 培養基中繼續培養六週之二段式培養法之植株再生率最佳,其植株再生率可 達 14.2%。 又於一段式葉塊培養法中培養基內植物生長調節劑組成, 則以含 2 ∼ 12mg/l BA 及 0.12 ∼ 1.2mg/l NAA 之 MS 培養基之葉塊培養植株再生率較佳,約 3.3 ∼ 8.3%, 相對地,含 2 ∼ 12mg/l kinetin 及 0.12 ∼ 1.2mg/l NAA 之 MS 培養基之植株再生率約 2.6 ∼ 4.1%,證實植物生長調節劑 BA 與 NAA 組合比 kinetin 與 NAA 組合之植株再生效 果佳,且其 C/A 比愈大則葉塊培植體之植株再生率亦愈高。 由葉塊培養再生出之小植株之 馴化成活率約 72%,且馴化出之植株與實生木瓜植株並無差異,由這些結果顯示,由葉塊培 植體再生出之木瓜植株之活性強。 |
| 英文摘要 | The leaf explants of 61-day-old papaya (Carica papaya var. Tainung No.5 ) were used as the materials of leaf disc culture for tissue. The leaf disc explants from papaya plants in which buds were removed could differentiate to plantlets, but there was no plantlet differentiated from leaf disc explants if the buds were not removed. The best percentage for the plantlet regeneration of leaf disc explants was observed when the 7th phyllotaxy at the 8th day after removing the buds, the plantlet regeneration rate was about 5.5% when cultured on the MS medium containing 2.2mg/l kinetin and 4.7 mg/l NAA. It showed that the removal of buds promotes the plantlet regeneration. When the one-step and two-step cultural methods were used to compare the plantlet regeneration rates after bud removal, the best one was the two-step cultural method which included culturing on the MS medium containing 2.2 mg/l kinetin and 4.7 mg/l NAA for two weeks and followed by transplanting to the plant growth regulators free MS medium for six weeks, the plantlet regeneration rate was 14.2%. The combinations of plant growth regulators in media influented the plantlet regeneration, the MS medium containing 2 ∼ 12 mg/l BA and 0.12 ∼ 1.2 mg/l NAA was better, with 3.3 ∼ 8.3% regeneration rates. However, the MS mediumcontaining 2 ∼ 12 mg/l kinetin and 0.12 ∼ 1.2 mg/l NAA showed only 2.6 ∼ 4.1%. These results confirmed that the combination of BA and NAA was better than the kinetin and NAA for the plantlet regeneration, and higher cytokinin/auxin ratios resulted in higher percentages of plantlet regeneration rates. The plantlets regenerated from leaf disc explants could be established in soil, an average survival rates of 72% was achieved after they had been acclimated. No phenotypic differences between the regenerated plantlets and the seedling plants could be noticed indicating that the vitality of the regenerated plantlets of papaya from leaf disc explants is as strong as seedlings. |
