中文摘要 | 電漿熔射生醫活性氫氧基磷灰石(HA)塗層披覆之鈦鋁釩(Ti-6A1-4V)合金植入 材料具有優良的生物相容性( Biocompatibility )與機械性質,於探討改善植入材料與骨 組織間固定( Fixation )效果之領域中,已引起廣泛地研究。本研究針對電漿熔射 HA 塗 層,就塗層之物理、化學特性以及塗層與骨組織間的生物反應等作探討。 在第一部份的實驗中,於鈦鋁釩合金基材上製備 HA 塗層,並研究塗層之特性。此部份結果 顯示 HA 塗層在顯微結構、相組成、結晶程度以及鈣磷莫耳比等塗層特性上, HA 塗層特性 為顯微結構緻密、雜相(三鈣磷酸鹽、四鈣磷酸鹽、氧化鈣)含量多、結晶程度指數低、鈣 磷莫耳比高。由於 HA 塗層之 OH 離子於電漿熔射後已大部份散失,因此 HA 塗層之相穩定 性比原始 HA 粉末差。 第二部份的實驗中,利用自行設計的中國銅錢植入模式( Chinese Coin Implant Moedl ) ,於狗之股骨( Femur )中,定量地評估 HA 塗層之生物反應。 所評估之生物反應包括引 導骨生能力( Osteoconductivity )和骨聚集能力( Osseointegration ),並分別以新 生骨癒合指數( NBHI )和新生骨附著指數( AI )代表之。 此部份結果顯示:4,6,與 12星期之植入後, HA 塗層所量測到的 NBHI 和 AI 值均較 Ti6A14V 合金高,且在統計上 顯著之差異。然而,於12星期植入後,HA塗層與Ti6Al4V合金其所量測之 NBHI 和 AI 值卻 明顯地降低。此原因可能由於骨重建時所引起。 |
英文摘要 | Revealing excellent biocompatibility and mechanical properties, plasma-sprayed bioactive hydoxyapatite (HA)-coated Ti alloy implants have raised much interest as an approach to achieve reliable implant-to-bone fixation. The aim of this study was to evaluate in vitro the physical and chemical characteristics and in vivo the biological responses of plasma-sprayed HA coatings (HACs). In phase I, three HACs on Ti-6Al-4V substrates were prepared using plasma spraying. The results demonstrate HAC is the higher in content of impurity phase, the least in index of crystallinity, and the higher in Ca/p molar ratio. Since the fact that the OH ion of HACs is lost almostly after plasma spraying, the phase stability of HACs is less stable than that of starting HA powders. By employing the Chinese Coin implant model in the cortex of canine femora, in phase II,the osteoconductivity and the ability of osseointegration of three HACs were assessed in terms of the new bone healing index (NBHI) and the apposition index (AI),respectively. Throughout all implant periods, the HA coated Ti6A14V implants revealed higher NBHI and AI than the uncoated ones, it appearing that the HA coated Ti6A14V was more osteoconductive than the Ti6A14V was. For HA coated implant, the evidence of direct bone-to-HAC contact. However, at the bone /Ti6A14V interface, there intervened a fibrous membrane without calcium content, indicating that Ti6A14V implant was not osseointegrated in the SEM field of view. The signs of partial dissolution of HAC within the remodelling canals were evident at the HAC-bone interface 12 weeks after implantation, accoungting for the slight decrease in NBHI and the obvious decrease in AI for HAC. |
