中文摘要 | 從鱒魚肝臟分離到的粒線體,具有高度外膜完整性(80-90%),此種粒線體被發現對ADP沒有反應性,當ADP添加於粒線體懸浮液中,並沒有增加粒線體的呼吸作用;然而,當把粒線體的外膜,以清潔劑Digitonin作選擇性地破壞,卻能恢復粒線體的ADP-dependent呼吸作用;或是在完整粒線體的懸浮液中,加入牛血清蛋白(Bovine Serum Albumin)(0.27%),亦能恢復粒線體的ADP-dependent呼吸作用。這個結果顯示粒線體外膜確是粒線體的屏障(barrier),利用外膜的屏障性來達到調解粒線體的功能。牛血清蛋白恢復粒線體的ADP-dependent呼吸作用,可能由於牛血清蛋白重新打開VDAC channel,而恢復其正常功能。 |
英文摘要 | Trout mitochondria were found to be unresponsive to adenosine 5'-diphos-phate (ADP) unless the outer membrane was damaged or bovine serum albumin (BSA) was added. Mitochondria isolated from trout liver with highly intact outer membranes (intactness 80 to 90%) showed no increase in respiration when ADP was added to the medium. However, ADP-dependent respiration was restored in the presence of 0.27% (w/v) of bovine serum albumin or triggered by the addition of same. The level of ADP-dependent respiration was inversely correlated with the degree of outer membrane intactness and was restored by treating intact mitochondria with digitonin. The results are consistent with the outer membrane being in a state that is poorly permeable to ADP and that the albumin relieves this permeabiltty barrier by opening voltage dependent anion-selective channel (VDAC) channels. |
