中文摘要 | 在本實驗中,體外培養的人類角膜內皮細胞經過含5mM,10mM,20mM,及25mM,的培養液培養兩天後,測試其角膜內皮細胞對葡萄糖攝取能力的改變及細胞cAMP濃度的變化。實驗結果顯示細胞對葡萄糖攝取能力 (nmole/min/mg protein) 會隨著培養液中所含葡萄糖濃度的增加而減少由0.18 (5mM),0.10 (10mM),0.07 (20mM) 降到0.06 (25mM),在經過胰島素處理過也發現葡萄糖攝取的能力由0.30 (5mM),0.11 (10mM),0.08 (20mM) 降到0.05 (25mM),同時經過高濃度的葡萄糖處理過的人類角膜內皮細胞不論是否經過胰島素處理過, 其細胞內cAMP的濃度(pmole/well)皆隨培養液內葡萄糖的濃度增加而減少,在胰島素處理組則由73 (5mM) 到20 (25mM),在無胰島素處理組由101 (5mM)到31 (25mM),而且經過胰島素處理過的細胞其cAMP的含量比沒有經過胰島素處理的細胞少,而且這兩組細胞葡萄糖的攝取有明顯減少趨勢,當細胞以1mM dibutyryl-cAMP培養一小時後,此減少現象很明顯的消失,由以上結果顯示,人類角膜內皮細胞若處在高糖份的狀況下會減少細胞對葡萄糖的攝取,而且這種改變可能經由cAMP的途徑。 |
英文摘要 | In this study, cultured human corneal endothelial cells were incubated in media containing various concentrations of glucose at 5mM, 10mM, 20mM and 25mM for 2 days. Then, the cellular 2-deoxyglucose uptake and camp concentation of cultured human corneal endothelial cells were measured. The results indicated that the activity of cellular glucose uptake of nmole/min/mg protein was decreased gradually from 0.18 (5mM), 0.10 (10mM), 0.07 (20mM) to 0.06 (25mM) after 2 days incubation with a high concentation of glucose. The glucose uptake in insulin-treated human corneal endothelial cells also exhibited a simlar declining effect in high glucose media from 0.30 (5mM), 0.11(10mM), 0.08 (20mM) to 0.05 (25mM), the cAMP concentration in human corneal endothelial ells was measured in the presence of high glucose media. It was indicated that the cAMP concentrations of pmole/well in both insulin-treated and non-insulin treated cells were also decreased after increasing the glucose concentration in the media from 73 (5mM) to 20 (25mM) and 101 (5mM) respectively. The cAMP concentration in insulin-treated cells was less than in non-insulin treated cells. This decreasing effect was significantly reversed by the addition of 1mM dibutyryl- cAMP to the cells for 1 hour in both groups. These results suggest that the diabetic state may decrease the 2-deoxygiucose uptake in human corneal endothelial cells via cAMP-dependent pathway. |
