中文摘要 | 本研究的目的即在篩選具有分解酚酸能力的細菌來解決土壤酚酸所引起的植物相 剋作用。 分別以 ferulic acid 及 p-hydroxybenzoic acid 為唯一碳源所篩選出來的分解 菌, 各別為 Bacillus sp. 486f 及 Rhizobium sp. 486p 菌種; 486f 菌對於 ferulic acid 100 ppm 之分解能力第 3 天殘餘百分率 45%,第 7 天殘餘百分率為 21%;486p 對於 p-hydroxybenzoic acid 100 ppm 之分解能力第 3 天殘餘百分率為 76%,在第 7 天殘餘百 分率為 51%,而 Pseudomonas putida 為 48%,P. putida 在第 7 天對之殘餘百分率為 0% 而 486p 之殘餘百分率僅為 51%,顯示 486f 其分解能力大於 P. putida。在酚酸的代謝實 驗中以 486f 及 486p 菌種對 50, 100, 1000 ppm 不同的濃度作為觀察, 發現低濃度下( 50 ppm )其代謝速率快,且完全分解; 高濃度( 1000 ppm )將造成抑制作用,而影響分 解作用。在添加葡萄糖作為外來碳源對酚酸分解作用的影響實驗中,結果顯示此分解菌能利 用葡萄糖作為外來碳源,且加速酚酸分解;若添加土壤溶液則不但加速分解,且分解完全, 結果顯示 486f 及 486p 之兩株分解菌都有胞內酚酸之存在,且 486f 胞內酚酸測定中發現 有 vanillic acid 之中間代謝產物的存在。 酚酸毒性試驗的結果顯示, 200 ppm 的 ferulic acid 已完全抑制萵苣的生長,而 p-hydroxybenzoic acid 其抑制率僅達 44.4%, 得知不同的植物毒物質其毒性將有差異。生物分析結果顯示,在接種分解菌後第一天種植萵 苣生長已經得到改善的效果。 |
英文摘要 | The purpose of this research is to solve the problem of allelopathy brought about by phenolic acids existing in the soil by bacteria that are capable of decomposing phenolic acids. This experiment separately employs. bacteria, Bacillus sp. 486f and Rhizobium sp. 486p that are isolated out by way of using ferulic acid and p-hydroxybenzoic acid as the carbon source. The remaining percentage of the decomposing capacity of Bacillus sp. 486f to 100ppm ferulic acid on the 3rd day is 45%, on the 7th day the remaining percentage is 21%; the decomposing capacity of Rhizobium sp. 486p to 100 ppm p-hydroxybenzoic acid has the remaining percentage of 76% on 3rd day, and the remaining percentage is 51% on the 7th day, but pseudomonas putida is 48%. The experiment inidicates the decomposing rate of P. putida to p-hydroxybenzoic acid on 7th day reaches 100%, while that of Rhizobium sp. 486p is only 49%. In the exsperiment using glucose as the carbon source to test influence of phenolic acid on the decomposition of phenolic acids, the result shows that these two bacteria can make use of glucose as carbon source and accelerate the decomposition, but through incomplete; The addition with soil solution in the experiment, it dose not only accelerates the decomposition of the phenolic acids, but also does the decomposition completely. The result of phenolic acid toxicity test indicates that ferulic acid completely suppress growth of lettuce at 200 ppm, and the rate of suppression of p-hydroxybenzoic acid only reaches 44.4%. The result of the experiment of bioassay shows that the growth of lettuce got improvement on the first day by receiving the bacteria treatment. |
