| 英文摘要 | Because infiltration of the middle ear submucosa by inflammatory cells is a constant feature of otitis media, and reactive oxygen species (ROS) are produced in large quantities by phagocytic cells and emerge as major inflammatory mediators. Using SV40-transformed Mongolian gerbil middle ear epithelial cells, we investigated if the predominant effect of activated inflammatry cells on middle ear epithelial cells was actually related to the release of oxidants. Polymorphonuclear leukocytes (PMN) were activated by N-formyl-methionyl-leucyl-phenylalanine (fMLP). Addition of activated PMN (10 �� /mL) to 10 �� /mL the basal compartment of the modified Ussing chamber resulted in a significant increase is Isc: 3.30 ± 0.45 μ A/cm ��, as compared with no increase for control (p<0.001). Catalase (10000 U/mL), which degrades hydrogen peroxide into water, deeply inhibited this process. This work demonstrated that reactive oxygen species are highly involved in the interaction between inflammatory cells and middle ear epithelial cells. This interaction might be a key in the impairment of mucociliary clearance and, thus, in the pathogenesis of otitis media. |
