中文摘要 | 上中下通用痛風丸(TFW)出 自金朝朱丹溪所著丹溪心法痛風篇,為一將各種辯證 如以綜合所設之方劑,臨床醫師常用於治療各類風濕病,然而其藥理與機轉之研究仍然闕如 。本研究以佐劑性關節炎之動物模型進行實驗,評估項目為關節腫脹度、關節組織病理檢查 ; 血液檢查則包括 albumin、ESR; 痛風丸對於角叉菜或組織胺誘發足蹠浮腫、鎮痛作用、 微血管通透性之影響,急性毒性試驗亦在相關所究之內。 結果顯示 TFW 無論對角叉菜足蹠 急性炎症或佐劑性關節炎之慢性炎症皆有明顯之抗浮腫作用, TFW(2g/kg) 抗角叉菜足蹠浮 腫效果與 Indomethacin (5 m/kg) 相當。皮膚視窗法發現 TFW(l g/kg) 能抑制組織胺引起 之微血管滲透性增加。TFW 對醋酸扭體及熱板致痛皆具顯著鎮痛作用。在佐劑性關節炎實驗 中, 關節組織病理發現痛風丸 (2g/kg) 及 Indomethacin 皆具減輕關節滑膜炎之作用。 TFW(l and 2g/kg) 對 ESR、 albumin 無影響, 而進一步評估自由機的傷害中, 並未發現 TFW(l and 2 gfkg) 各組與控制組比較,血漿 SOD 活性、LPO 的濃度、肝組織內 SOD、GSH -PX 活性與 LPO 的濃度並未有統計上之差異。 由以上結果顯示,上中下通用痛風丸有其一 定的抗炎止痛效果,且在有效劑量下未有明顯毒性,可望在抗炎鎮痛方面提供另一選擇。至 於 TFW 治療關節炎的機轉,則有待進一步研究。 |
英文摘要 | Tonq-Feng-Wan (TFW ) is a traditional chinese herbal remedy having heat -clearing, cold -warming, dampness transforming, hemodynamic, and analgesic effects. It has been Used by traditional Chinese physicians for the treatment of rheumatic disorders, such as rheumatoid arthritis, gouty arthritiS, fibromyagia. However, the scientie studies of this recipe for rheumatoid arthritis is still in want. Adjuvant - induced arthritis (AIA ) was used in this study as a animal model of rheumatoid arthritis ESR and serum albumin were checked also. Additionally, carrageen-induced hindpaw edema (CIE) , plasma extravasation induced by inflammatoy materials (histamine, 5-HT, bradykin, PGE2) in skin-window method, histamine-induced hindpaw edema, acetic acid induced writhing pain, and hot plate pain model, were included in this study. The following results were obtained: Foot edema of AIA could be suppressed by TFW (Ig, 2g/kg) significantly during the whole course. However, indomethacin was only effective in first stage This finding was supported by pathological picture. TFW had no effects on ESR and Albumin. TFW(2g/kg) and indomethacin (5mg/kg) both suppressed CIE to the same degree, TFW could suppress the extravasation induced~by histamine. The analgesic effects of TFW was proved by wirthing reponse and hot plate method. The evaluation of TFW against free radical-induced toxicity, including activity of SOD in plasma and liver tissue, GSH-PX in liver tissue, and LPO in plasma and liver tissue, was shown statistically to be no significant difference between drug treated animals and control. Accordingly, TFW has definite antirheumatic and analgesic effects, and otherwise, no apparent acute toxicity was noted under effective dosage. It may become a alternate choice in antirheumatoid drug. However, the mechanisms of TFW on immunologic aspects need to be further investigated. |
