中文摘要 | 本文乃比較麩氨基硫S-轉移�t(GST-π)在人類正常口腔組織、口腔癌、血漿及各種不同細胞株中之含量,以做為口腔腫瘤標誌之參考。GST-π濃度的測量都是利用自製並定性之單株抗體,以ELISA方法來測量及定量。實驗結果,從口腔癌中測出GST-π平均值則為42.23±29.00μg/mg protein,其腫瘤周圍正常組織之GST-π平均值則為21.36±16.73μg/mg protein,此結果與患者年齡或腫瘤期別並無關連。從細胞株中,測出人類口腔癌、肝癌及金田鼠口腔癌之GST-π濃度平均值分別為61.64μg/mg protein、65.35μg/mg protein,及55.47μg/mg protein,此結果比人類正常口腔纖維細胞之GST-π平均值17.19μg/mg protein高出許多。而GTS-π在口腔癌患者之血漿中也比正常人高出許多,第一、二期之患者比正常人高出約46,而第三、四期之患者則高出約55。研究結果也發現,患者是否咀嚼檳榔與血漿中之GST-π濃度沒有直接關係。歸納以上結果顯示GST-π含量無論在口腔癌患者之腫瘤或血漿中都比正常人來得高,GST-π可以考慮作為輔助早期診斷之工具。 |
英文摘要 | The purposes of this study were to evaluate the clinical usefulness of glutathione S-transferase-π (GST-π) as a tumor marker in patients with oralcancer. The GST-π content was measured with a sandwich enzyme-link immunoassy (ELISA) system. The GST-π in oral cancer tissue (42.24 ± 29.40μg/mg protein) is higher than nsurrounding normal tissue (21.36 ± 16.73 μg/mg protein). There is no relation with age or stage of the patients. The GST-π in three carcinoma cell lines OC2, HA-22T, and H ranged from 55 to 65 μg/mg protein. Normal fibrob last cellscontained 17.19 μg /mg protein. The GST-π in the plasma of oral cancer patients (34 ± 27.43 ng/ml) is higher than in normal healthy persons (14.00 ±9.16 ng/ml). 55of stage III and stage IV oral cancer patients and 46 of those with stage I and IIhad elevated levels of GST-πin the plasma. The GST-π level was not co-related with betel nut chewing. In conclusion, GST-π is considered to be a good diagnostic aid for early detection of oral cancer patients. |
