中文摘要 | Histone H3 基因是一種與細胞增殖有關的基因,而細胸增殖與腫瘤的診斷、分 期及治療計畫有著密切的關聯。本研究收集中山醫學院牙科組織病理確認為牙齦癌之標本 10 例為實驗組,及正常牙齦組織 4 例作為對照組,以進行研究分析。切片厚度為 8 μ m 、以原位雜交法( in situ hybridization ), 利用□ S 標定 H3 mRNA 來偵測切片中細 胞複製活性之表現。結果顯示,正常牙齦組織切片中,僅在正常牙齦的黏膜上皮其基底層細 胞偶有 H3 mRNA 之訊號產生;在牙齦癌切片上,發現絕大部分癌細胞病變區有顯著 H3 mRNA 訊號。綜合本研究顯示,H3 mRNA 之表現,可為牙齦癌腫瘤茩 M 增殖之良好指標。 |
英文摘要 | Histone H3 is a proliferation-dependent gene. Cancer is a disease involving inappropriate cell division. A realistic model is greatly needed to understand the biology of altered proliferation in cancer as compared to normal tissue and to use information on proliferation capacity as a basis of cancer prognosis and treatment. In this study, the expression of the histone H3 gene was analyzed by in situ hybridization (ISH) to determine the proliferation patterns of malignant gingival lesions. Samples f four normal gingivae and ten gingival carcinomas were obtained from Chung Shan Medical and Dental College Hospital. Eight- μ m sections were cut, mounted onto gelatin coated slides and processed by the standard method for ISH with □ S-labeled H3 riboprobe. The H3 mRNA hybridization signals were in accordance with the histopathology of the biopsies. In normal gingival, H3 mRNA signals were only seen occasionally in the basal layer. In gingival carcinomas highly elevated H3 mRNA expression was noted. In the present study, the sensitive nature of the histone H3 gene could be used as an indicator of proliferation in normal and malignant gingival lesions. It might be important in determining subsequent treatment modalities for oral cancers. |
