中文摘要 | 豬紅血球在酸性狀態添加可溶性羧甲基纖維素鈉 (sodium carboxymethyl cellulose,Na-CMC)分離血球蛋白與原血紅素(heme)。原血紅素中鐵的組態究屬二價或三價,實關係原血紅素做為有機鐵源的價值。採用I,1O-o-phenanthroline,Ferrozine 即 (3-(2pyridyl)-5,6-bis(4-phenylsulfonicacid)-1,2,4-triazine)及赤血鹽(potassium ferricyanide)等二價鐵試劑與以 Na-CMC 分離之血球蛋白、原血紅素洗液之反應,結果發現1,10-o-phenanthroline 與 Ferronine試劑發色穩定,血球蛋白與原血紅素洗液添加上述兩種試劑後之吸光值增加。赤血鹽試劑與高濃度亞鐵溶液反應可立即由草綠變藍色並有懸浮沉澱。於低濃度下,反應進程極慢。血球蛋白與原血紅素洗液與赤血鹽反應在 1~2 天後有少許藍色沉澱。由上結果驗證,血球蛋白與原血紅素洗液在酸性下亦保有二價鐵之組態。 |
英文摘要 | There were few reports to elucidate the confirmation of iron in acid hydrolysis of animal blood globin and heine eluents. By the addition of sodium carboxymethyl cellulose at pH 2.0 to red blood cell concentrate, globin and heme eluents of porcine hemoglobin were separated. The reagents 1, 10-o-phenanthroline, ferrozine, and potassium ferricyanide were used to determine the presence of ferrous iron in globin and heme eluents. After reaction with 1, 10-o-phenanthroline and ferrozine, the color of blood globin and heine eluents was slightly changed, and the absorbance of those eluents at 510 or 562nm was also increased. The reaction between potassium ferricyanide and blood globin or heine eluents was very slow, but some blue precipitates could be observed after 1~2 days. Based on the observation that the absorbance increased after adding these ferrous reagents, ferrous iron in acid hydrolysis of blood globin and heine eluents could be detected. |
