中文摘要 | 本研究之目的在探討大白鼠初代肝細胞於0.5mM油酸(oleic acid,18:1)、亞麻油酸(linoleic acid,18:2)或次亞麻油酸(α-linolenic acid,18:3)培養下,該細胞之生存力(以lactate dehydrogenase leakage為指標,LDH leakage)、脂質過氧化(以thiobarbituric acid reactive substances為指標,TBARS)、麩胱甘呔(glutathione,GSH)與其代謝相關酵素活性含量之影響。結果顯示:18:2及18:3組之細胞LDH leakage顯著較控制組高。18沼組之TBARS值顯著增加,而18:1組則無顯著影響。細胞內總GSH含量在18:2及18:3各組均顯著較控制組低;18:3組其GSH/GSSG比值顯著較其他組低。各脂肪酸組glutathione peroxidase(GSHPx)活性,隨著培養時間增加而有下降之趨勢,18:2及18:3組顯著較控制組低;glutathione reductase(GSHRd)活性,在培養至第24小時時,18:3組顯著低於控制組。Glutathione S-transferase(GST)隨著培養時間增加而有下降之趨勢,在培養至第24小時時,除了18八組外,其他組均顯著較控制組低。綜合上述,肝細胞以18:3處理時,因脂肪酸的雙鍵數目較多而使細胞之生存力降低,發生脂質過氧化現象明顯增加。 |
英文摘要 | The purpose of this study was to investigate the effect of 0.5 mM leic acid(18:1), linoleic acid (18:2) or linolenic acid (18:3) on cell viability (determined asthe leakage of lactate dehydrogenase, LDH leakage), lipid peroxidation (measuredas thiobarbituric acid reactive substances, TEARS), glutathione (GSH)content andenzyme activities related to GSH metabolism in primary rat hepatocytes. Thedata showed that the LDH leakage of 18:2 and 18:3 groups was significantlyhigher than the control (without unsaturated fatty acid addition). The TEARSconcentration of the 18:3 group was significantly higher than the control; however, there was no significant effect found in the 18:1 group. The total intracellular GSH levels of 18:2 and 18:3 groups were significantly lower than thecontrol; the GSH/GSSG ratios of 18:3 group was significantly lower than theother groups. The glutathione peroxidase (GSH Px) activity decreased withincreased time of incubation. GSH Px activities in cells treated with 18:2 and 18:3groups were significantly lower than the control. After 24 hours of incubation,the glutathione reductase (GSH Rd) activities for 18:3 group were significantly lower than the control. The glutathione S-transferase (GST) activity decreasedwith the increase of incubation time; thus after 24 hours of incubation, the GST activities of 18:2 and 18:3 groups were significantly lower than the control. In conclusion, when the hepatocytes were treated with 18:3, the cell viability wasreduced and the lipid peroxidation ncreased, probably because 18:3 has moredouble-bonds than 18:1 and 18:2. |
