中文摘要 | 本試驗目的在於探討飼料中色胺酸測定方法。以單味飼料原料之魚粉、大豆粕、肉骨粉、血粉、葵仔粕、啤酒粕、番茄渣、狼尾草及玉米等為材料,經6N NaOH鹼水解後,分別以比色法及高效液相層析(HPLC)螢光檢測法定量色胺酸之量。比色法是使色胺酸與4-dimethylamino benzaldehyde(DMAB)液反應後呈色之定量法;HPLC螢光檢測則利用層析分離後色胺酸之螢光性寔量之。分析結果,比色法及HPLC螢光檢測法分析結果在樣品內重複性之變異係數分別為11.56%及4.17%,以比色法分析變異較大;回收率則分別為0~105%及90~100%,HPLC法較為穩定。HPLC螢光法較快速、穩定,適合於作為實驗室例行性檢驗方法。 |
英文摘要 | The objective of this experiment was to establish the method for determination of tryptophan in feeds. The feed ingredients used were fish meal, soybean meal, corn, blood meal, brewer's pulp, sunflower meal, meat and bone meal, tomato pomace, and napier grass. Protein in feeds were hydrolysed with 6 N sodium hydroxide solution and followed by reacting with p-dimethylamino benzaldehyde (DMAB) for colorimetric method or by high-performance liquid chromatography with the fluorescence detector. Comparison of the two methods showed that the variation of tryptophan contents in feeds was larger for colorimetric method than that of HPLC procedure (11.56% vs. 4.17%). The recovery rate were 0~105% vs. 90~100%. The advantage of using HPLC were rapid, sensitive and specific. The results suggest that using HPLC for determination of tryptophan in feed is recommended for routine analytical use. |
