中文摘要 | 本實驗之目的在於建立一種既簡單快速又可連續檢測五種quinolone 類抗菌 劑:enrofloxacin, flumequine, nalidixic acid, oxolinic anid及sarafloxacin 之方 法。實驗結果顯示以裝有前置分離管之LiChrospher RP-l8分離管,配合0.01 M流動相 oxalic acid-acetonitrile-methanol-tetrahydrofuran(7.5:1.5:0.5:0.5,pH=3.0) ,可清 晰地將上述五種quinolone 類藥物分離。 以水和acetonitrile(1:4)為萃取液時,回收率 可高達68.3-84.9%。 以26O nm 波長之紫外線偵測器同步檢測肌肉中上述五種 quinolone類藥物時,其最低檢測濃度可達0.07-05 μg/g,並且分析一個樣品時間平均僅 需四十分鐘。 |
英文摘要 | The aim of the study was to develop a simple and rapid method for the simultaneous determination of residual quinolones; enrofloxacin, flumequine, nalidixic acid, oxolinic acid, and saraflowacin by high performance liquid chromatography (HPLC). The method was carried out on a LiChrospher RP-18 column (150 x 4.6 mm I.D.) with 0.01 M oxalic acid- acetonitrile- methanol- tetrahydrofuran (7.5:1.5:0.5:0.5; pH 3.0) as the mobile phase. UV-VIS detector was used at 260 nm wavelength. The results showed that the recoveries of quinolones extracted with water: acetonitrile (1:4) from eel's muscle were 68.3-84.9%. The detection limits of quinolnes by our method were 0.07-0.5 ppm. The analytical time per sample was less than 40 min. Thus, our results shows that a simple, sensitive and rapid HPLC may be a feasible alternative tool for detecting quinolones / fluoroquinolone in animal tissues. |
