英文摘要 | Cultures were initiated from nodal explants. Explants grown on a solid medium containing costituents of MS formula, naphthalene acetic acid at 0.01 mg/l, and benzyladenine at 1 to 10 mg/l, under photosynthetic photon flux density (PPFD) at 80 μ mol/m �� sec for 4 to 5 weeks, produced the best lateral shoots for subculture. A PPFD at 40 μ mol/m �� sec was optimum for the growth of fresh and dry weight of 'Pitica' explants. However, a PPFD at 20 μ mol/m �� sec stimulated multiplication. 'Samantha' explants got higher fresh weight and more multiple shoots under a PPFD at 25 μ mol/m �� sec. 'Landora' explants got higher fresh weight under a PPFD at 40 μ mol/m �� sec. The CO �� concentration in vessels decreased during the explants cultured under lighting. However, after culture for 4 weeks, the CO �� concentration in the sealed vessels where explants growing on a medium containing sucrose was high to 8877 ppm at the beginning of lighting. Moreover, at the end of 16-hour lighting the CO �� concentration was still high to 6041 ppm. The dry weight of 'Samantha' explants was heavier under CO �� enrichment by adding CO �� to 1000 ppm twice a day. |
