英文摘要 | 隨機擴增多型性 DNA(RAPD) 標誌被用於臺灣金線蓮及高雄金線蓮品種的鑑定上。 此二品種形態上最主要的差別在於臺灣金線蓮的花部唇瓣上具有梳狀構造,高雄金線蓮則無 。 RAPD 標誌為金線蓮品種的鑑定,提供了一個既快速又簡便的工具。本研究即針對該目的 進行四十條引子的篩選,結果分別得到由四條引子所產生的七個 RAPD 標誌,具有品種的專 一性,其中三個標誌為臺灣金線蓮所特有,而其餘四個標誌則為高雄金線蓮所特有。為了評 估這些標誌的再現性,本研究同時進行一組改變模板 DNA 濃度的 PCR 反應,結果在 25 微 毫升的反應體積中,DNA 含量從 5ng 至 40ng,這些標誌均能穩定出現。本研究的 RAPD 標 誌不但可應用於金線種品種的篩選,更有利於今後植物育種之用。 d, and seven species-specific RAPD markers generated from polymerase chain reaction (PCR) with four random primers were obtained. Of the markers, three were specific to A. formosanus and four to A. koshunensis. To evaluate the reproducibility of these RAPD markers, amplification reactions were performed in various template DNA amounts. The markers were highly conserved in the reactions in which DNA content ranged from 5ng to 40ng in 25 μ L reaction volume. The RAPD markers can be applied both to identification of Anoectochius species and to assessment of the extent of hybridization in hybrids between A. formosanus and A. koshunensis, information about which facilitates the breeding program process. |
