| 中文摘要 | 血管內皮細胞構成血管之內壁,直接承受因血液流動所產生之血液動力 學作用力並作適當調節,醫學研究顯示此種作用若發生異常,則可能導致動脈硬 化等血管病變,本文探討以體外實驗方式,使用流動或伸張裝置以瞭解兩種血液 動力學作用力,即剪力與張力對人類血管內皮細胞之影響,包括細胞形態、基因 表現、蛋白質生成、胞內訊號傳遞途徑等,研究顯示細胞功能除受化學刺激因子 如荷爾蒙、養份等調控之外,物理作用力如剪力與張力亦能顯著改變細胞生理現 象,這些發現有助於深入瞭解何以血管病變與血液動力學之間有密切關連,亦可 協助心臟血管疾病之研究、治療、乃至預防。 |
| 英文摘要 | Vascular endothclial cells form a lining of blood vessels and constantly experience hemodynamic forces generated by the blood flow. The abnormality in theresponse of endothelial cells to hemodynamic forces may result in vascular diseasessuch as atherosclerosis. This article discusses the effects of hemodynamic forces,including shear stress and cyclic strain, on cultured human endothelial cells. Most ofthe data was obtained from in vitro experiments using either flow chambers or strainchambers. Shear stress or cyclic strain was found to change the morphology, geneexpression, protein production, and intracellular second messengers in endothelialcells. Therefore, cell functions are affected not only by chemical stimulators likehormones and nutrients but also by mechanical forces like shear and strain. Thesefindings will illuminate the hemodynamic aspect of vascular disease and also contribute to the prevention and therapy of cardiovascular disorders. |
