中文摘要 | 褐藻酸鈉及羧甲基纖維素鈉可沉澱回收豬血漿蛋白。本研究係改變反應pH及蛋白質與多醣類混合比混(w/w),探討二反應因子對回收率之影響及較佳加工條件。結果顯示,二加工因子均對回收率有顯著(p<0.01)影響,由單因子試驗設計所得結果知,以褐藻酸鈉操作時,在pH3.5~4.0、混合比例(蛋白質:多醣類)4:1~6:1能獲得較高之回收率;以羧甲基纖維素鈉捆作時,在pH4.0~4.2、混合比例2:1~4:1能獲得較高之回收率。由多因子試驗設計所得結果知,以褐藻酸鈉操作時,當pH3.7、混合比例5.8:1~6.2:1時,其回收率接近100%,以羧甲基纖維素鈉操作時,當pH4.0、混合比例4.2:1時,其回收率達97.4%。 |
英文摘要 | This study was conducted to investigate the feasibility of protein recovery from pig blood plasma by precipitation with sodium alginate or sodium carboxymethyl cellulose. The optimum pH and mixing ratio (w/w) of protein to polysaccharide on recovery were studied. Results showed that pig blood plasma protein was effectively precipitated by soduim alginate and sodium carboxymethyl cellulose, and that the rcovery of plasma protein was significantly (p<0.01) affected by the pH and mixing ratio (w/w) of protein to polysaccharide. In one-factor-at-a-time experiments, higher recovery was observed when experiments were conducted at pH 3.5~4.0, mixing ratio (protein: polysaccharide) 4:1~6:1, and at pH 4.0~4.2, mixing ratio 2:1~4:1 with sodium alginated and soduim carboxymethyl cellulose, individually. Results from the two-factor factorial design showed that the highest recovey was 97.4% by precipitation with sodium coarboxymethyl cellulose at pH 4.0, mixing ratio 4.2:1 and approximately 100% by sodium alginate at pH 3.7, mixing ratio 5.8:1~6.2:1. |
