中文摘要 | 本報告探討以鴨B型肝炎病毒(duck hepatitis B virus,DHBV,帶原鴨做為篩 選具抗親肝DNA病毒(hepadnavirus)藥物之動物模式之可行性。用6隻1週齡DHBV 陽性雛菜鴨每隔3天以點狀雜交法 (Dot-blothybridi zation)測定其血中DHBV DNA 濃度至6月齡,結果得知此期間內其血清DHBV DNA平均濃度在27.76±3.39至 114.73±42.56pg/10ul間,但大部份在40±10pg/10ul。 DHBV帶原鴨從37日齡至187日齡測定其血清中二種肝源性酵素:氨基丙酸轉氨醣 (alanine aminotransferase,ALT)及天門冬酸鹽轉氨�t (aspartatetransferase,AST)之活 性值,結果顯示在試驗期間並無顯著變異,DHBV帶原鴨血清中ALT及AST活性 值與DHBV陰性仍之結果並無顯著差異。2隻DHBV帶原鴨各經6次肝臟生檢術取 出肝臟組織做組織病理學及病毒學分析,組織病理學檢查結果並無慢性活動性肝 炎或慢性持續性肝炎等病變。病毒學分析結果,可分析relaxed circular(RC)型, covalently closed circular(CCC)型及single strand(SS)型等DHBV DNA分子之複製中間 體。 由以上結果言,自然感染DHBV帶原鴨適合做為篩選具抗親肝DNA病毒效力藥物 之動物模式及探討藥物之毒性和抗病毒作用機轉。 |
英文摘要 | The possibility of duck hepatitis virus (DHBV) carriers as ananimal model for screening anti-hepadnavirus drugs was studied. The fluctuation of DHBV-DNA concentration in sera of 6 ducksfrom one-week to 6-month-old was determined every 3 days withDot-blot hybridization technique. The result showed that the averagevalues were between 27.76±3.39 and 114.73± 42.56pg/10ul, but most ofthen were at 40±10pg/10ul. The activities of two liver enzymes of asparate aminotransferase(AST) and alanine aminotransferase (ALT) in DHBV- carriers weredetermined from 37-day-old duckling to 187-day-old ducks. The resultsshowed that there were no significant variations during the testperiod. The hepatic enzymes of ALT and AST were no obviousdifferences between the DHBV positive and negative ducks. The liver was biopsied 6 times in each of the two DHBV carrierducks for histologic examination. No chronic active or persistenthepatitis lesions was found in the liver biopsies. The relaxed circular, covalently closed circular and single strandforms of the replication intermediate of DHBV could be detected inthe hepatocytes from the liver biopsies. Based on the results mentioned above, spontaneous DHBV carriersmay present a suitable animal model for screening drugs withanti-hepadnavirus potencies and for additional studies of theanti-viral activity mechanism and the toxicity of the drug. |
