| 中文摘要 | 利用胡蘿蔔Ri T-DNA轉型根及番茄根雙相培養均能於試管內生產囊叢 枝茵根菌Gigaspora gigantea孢子,且以番茄根培養者產孢量較高,於直徑9公分培 養皿上可生產72粒孢子。培養新生孢子能當作接種源於試管內再接種,而培養根 段亦可繼代培養,並順利產孢,並可用於盆栽土來大量生產接種源。而繡球屬 Glomusmosseae 的雙相培養產孢較困難。由於雙相培養僅產生少量無茵孢子,且 根段菌根感染率亦低(20),顯示雙相培養技術並不適合大量生產無菌接種源以供 試管內接種,卻適用於菌根生理研究、型態觀察及作為純系接種源之用。 |
| 英文摘要 | /a v/tro sporulation from dual culture of Ri T-DNA transfromedcarrot root and tomato root with vesicular-arbuscular mycorrhizal fungus (VAMF) Gigaspora gigantea were established, and amongthese treamants, best sporulation (approximately 72 spores in each 9cm petri dish) was consistently observed in that cultured withtomato root segments. The newly-formed spores could be used asinocula for next dual cultures, while the cultured root pieces couldbe used for subculture, as well as the mass production of pure lineinoculum. Attempt of dual culturing Glomus mosseae was notsuccessful, and only few sporulation were obseved, on which moreworks should be done. The results indicated that the dual culturetechnique was not suitable for mass production of axenic VAMFinoculum due to the low sporulation level; however it could be usedfor physiological studies, morphological observation, as well as pureline inoculum production. |
