中文摘要 | 由植物根圈及土壤中篩選出之細菌Bacillus subtilis,可利用有機氮源與不同 之碳源同時產生蛋白質分解酵素與澱粉分解酵素。 每公升培養基內含 8.0g 葡萄糖,10.0g tryptose,此細菌在 pH7.0,30 ℃及 125rpm 培養 21 小時, 蛋白質分解酵素之活性最高 ( 1300 CDU/ml ), 但隨著細菌進入產孢階段,則活性下降,培養 27 小時後之澱粉分解 酵素活性最高( 195 mg/min . g ),此活性一直維持到孢子釋出時才稍下降。 研究結果 亦顯示,麩皮、豆粉與魚精粉為相當好之氮源,以其取代培養基中之 tryptose 時,不僅提 高蛋白質分解酵素與澱粉分解酵素之活性(分別為 2500 CDU/ml 與 348 mg/min . g ), 且有效降低了培養基之價格。此菌株產生之蛋白質分解酵素對熱敏感,但其所產生之澱粉分 解酵素為耐熱性的。細菌孢子回收後,再令其發芽接種時,仍可維持相同的酵素活性,顯示 此等酵素之生產相當穩定。 |
英文摘要 | The beacterium, Bacillus subtilis, isolated from rhizosphere soil, produced both protease and amylase when it was cultured with an organic nitrogen source and different carbohydrates as a carbon substrate. Maximum protease activity (1300 CDU/ml) was obtained after 21 hours when the bacterium was cultured under optimal conditions (pH 7.0, 30 ℃, 125rpm), however, this enzyme activity declined when cells started to sporulate, maximum amylase activity (195 mg/min . g) was obtained after 27 hours cultivation, this activity didn't decay until spores were released. When tryptose was replaced by wheat bran, soybean meal or fish soluble, protease and amylase activity was 2500 CDU and 348 mg/min . g, respectively, indicating that cheaper and better nitrogen sources could effectively reduce the cost of spore production. The protease produced by this strain is heat-labile, whereas the amylase is heat stable. |
