中文摘要 | 本試驗乃以五種種間雜交種百合為材料,觀察花粉母細胞之減數分裂行 為及其花粉活力。亞洲型雜交種百合 "Revival"花粉母細胞之減數分裂極為正 常,在第一減數分裂中期每細胞平均之染色體配對為11.9II+0.2I,每一花粉母細 胞在第二減數分裂四分子期形成四個單元子細胞,所形成之花粉稔性高,產生之 2N花粉頻率低於1以下。種間雜交種79418-2及79418-7(此二者乃同由 LiliumX"Enchantment"與L.pumilum雜交選出之不同雜交種)在第一減數分裂中期亦 顯現高頻率之二價體,平均每細胞含二價染色體分別為11.9+I及11.9+I,然而不規 則之減數分裂,二價體非導向赤道板,不平衡之染色體分離及染色分體片段形成 而造成低的花粉稔性,此二雜交種之2N花粉形成機制在於無第二減數分裂,2N 花粉約佔28,此結果與減數分裂觀察在四分子期中產生二分子之機率約一致。種 間雜交桓89356-1與89352-6(此二者乃同由L.longiforum與L.henryi雜交選出之不同 雜交種)在第一減數分裂中期顯現極低的配對頻率,二價染色體分別為1.6II及 2.6II,不正常之減數分裂造成極低的花粉稔性(0-2.3),2N花粉形成之機制或由於 在第一減數分裂後期之子染色體分離。 |
英文摘要 | Meiotic observations in PMCs and pollen viability were made in five interspecific hybrids of Lilium. Asiatic hybrid 'Revival' ((L. X 'Eurovision') X (L. X'Rosefire')) showed the quite normal meiotic division. The mean chromosome pairingfrequency is 11.9II + 0.2 I per cell at metaphase 1. Four haploid cells wereformed in each PMC at tetrad stage of the second meiotic division. This hybridshowed a high pollen fertility and a low frequency of 2n-pollen (below 1). Hybrids 79418-2 and 79418-7 (different hybrids from the same cross (L. X 'Enchantments') X (L. X pumillim)) also showed very high bivalent frequency (11.9* IIand 11.9' II respectively) at metaphase-I. However, irregular meiotic divisions including bivalents un-oriented on the metaphase plate, unbalanced chromosome separation and chromatid fragmentation resulted in low pollen fertility. The mechanismof 2N-poUen formation of these two hybrids rises from the omission of the seconddivision. The percentage 2N-poUen formation was estimated on 28 using flow cytometry. This was in agreement with the percentage dyad formation in tetrad stage.Hybrids 89356-1 and 89352-6 (different hybrids from the same cross L. longiflorumX L. henryi) showed very low pairing frequency (1.6 II and 2.6 II respectively).Abnormal meiotic division resulted in extremely low pollen fertility (0 to 2.3). Themechanism of 2N-pollen formation of these two hybrids probably rises from abnormal chromatid separation at telophase-I. |
