中文摘要 | 為探討薄層層析─火焰離子化檢出儀在定量分析飼料中磺胺劑之適用性,乃進行本實驗。由薄層層析法所得多種磺胺劑之分析結果發現,為了避免飼料中其他所合成分之干擾,Rf=0.50以下的磺胺劑方能適用 (展開液為氯仿/第三了醇 (85/15, v/v) )。在以空白飼料添加已知含量之Sulfathiazole (Rf= 0.30) 方式,所求得之回收率為110%。以薄層層析─火焰離子化檢出儀定量分析飼料中Sulfathiazole (Sulfaguanidine為內部標準品)含量,所得之線性回歸方程式為Y= 1.3652X-0.3193,r=0.9820。對於已知含量Sufathiazole(100ppm及10ppm)所進行之分析結果發現,以一般的液相層析儀所得之分析值為91.4ppm及8.8ppm,以薄層層析火焰離子化檢出儀所得之分析值則為120.5ppm及11.6ppm。雖然利用薄層層析─火焰離子化檢出法所採取的方法較為簡便,然而自於其層析管柱的理論板數較低;在部分雜質的影響下,使得利用薄層層析─火焰離子化檢出法所得的分析值略大於液相層析法所得者。 |
英文摘要 | The objective of this study was to understand the applicability and the content of sulfonamides from feed by thin layer chromatographyflame ionization detector (TLC-FID). Base on the method of thin layer chromatography (TLC) by using chloroform: tert-butanol (v/v) as developing system, the results indicate Rf value of sulfonamides were lower than 0.50 and could be quantified by TLC-FID. The recovery efficiency of sulfathiazolefrom blank feed with acetonitrile was 110% directly. The linear relativity equation of sulfa-thiazole quantifed from feed, and suIfa-guanidine by the method of TLC-FID as internal standard, is shown y = 1.3562x-0.3193, r = 0.9820. the results of the 100 and 10 ppm suIfa-thiazole from feed quantified by HPLC were 91.4 and 8.8ppm, respectively. However the results quantified by TLC-FID were 120.5 and 11.6ppm, respectively. The data analyzed have shown that the method of TLC- FID would be easily applied to quantify sulfonamides from feed. Under the influence of partial impurity, the results quantifed by TLC-FID were higher than that quantified by HPLC. |
