中文摘要 | 本試驗採用C₄型植物小馬唐、千金子與鼠尾栗為主要材料,此三種植物依其脫羧作用主要酵素,分屬NADP-ME、NAD-ME和PCK生化亞型,且以往均未曾被報告為C₄型植物。試驗係針對葉片粗萃取液,分析三種脫羧酵素的特性,供為進行C₄亞型歸類之依據。試驗結果發現鼠尾栗PCK及玉米NADP-ME酵素並不具明顯之光活化特性;NADP-ME(小馬唐)與PCK在萃取後八小時仍能維持高的酵素活性,NAD-ME(千金子)活性則於萃取後逐漸降低。NADP-ME、NAD-ME與PCK均為41℃。在三個酵素的受質Km值方面,NADP-ME為1.027mM malate,PCK為0.096mM OAA與0.082mM ATP。綜合對三種新發現為C₄型植物脫羧酵素進行各項分析的結果,認為與其他同亞型植物的酵素具有相似特性。 |
英文摘要 | Three C₄ acid decarboxylating enzymes of C₄ pathway photosynthesis, NADP-malic enzyme (NADP-ME), NAD-ME and phosphoenolpyruvate carboxykinase (PCK) were studied from their in vitro characteristics from leaf crude extracts of, respectively, Digitaria radicosa (Presl) Miq. (NADP-ME subtype), Leptochloa chinensis (L.) Nees (NADP-ME subtype). And Sporobolus fertilis (Steud.) S. D. Clayton (PCK subtype). The 3 weed species have not been reported previously to possess C₄ pathway photosynthesis. Experimental results indicated that activity of PCK did not show significant diurnal response. Optimal assay pH for NADP-ME, NAD-ME and PCK were 8.6, 7.2 and 7.8, respectively. Optimal temperature was 51°C for NADP-ME, and 41°C for both NAD-ME, and PCK. Substr5ate Km values were 1.027 mM malate for NADP-ME, and 0.096 mM OAA and 0.082mM ATP for PCK. It is suggested that characteristics of the 3 enzymes are similar to those of the corresponding enzymes from other species. |
