| 中文摘要 | 本研究主要利用核酸限制酶切片長度多形性的技術探討人類磷酸果糖激酶(PFK)基因5'端的核酸序列之保留性。從三十個人體白血球及五種人類癌細胞株中抽取DNA,以人類PFK基因5'端的2.5Kb染色體DNA片段,兔肌PFK基因的3.5Kb與8.9Kb染色體DNA片段,以及人類肝臟型PFKcDNA等四種DNA為探針,進行核酸雜交實驗,結果發現僅人類PFK基因5'端的2.5Kb染色體DNA片段可偵測到雜交訊號。在30位受試者中,選用BamHI,EcoRI,HaeIII,HhaI,HindIII,KpnI,MspI,PstI及SalI等九種核酸限制酶檢測並未顯現PFK基因的多形性,因此推論人類PFK基因5'端的DNA序列在基因演化上具有相當強的保留性,此可能與保留PFK的催化活性有關。其中只有一例外:一位高血脂病人,其BamHI及PstI處理後所得PFK基因的RFLP與其他人不同,此可能為人群中之特例,原因有待進一步研究。在五個人類癌細胞株的研究中,其中四株得到與正常人相同的結果;另外一株TSGH細胞株則顯現不同的結果,其原因可能是此細胞株分化較差及染色體不穩定所表現的結果。 |
| 英文摘要 | The purpose of this study is to investigate the genetic conservation at the 5' region of human phosphofructokinase (PFK) gene by using RFLP (restriction fragment length polymorphism) analysis. DNA was isolated from human lymphocytes and analyzed by using various 32P-labeled probes including HuPFK-2.5Kb genomic fragment, RmPFK-3.5Kb and -8.9 Kb genomic fragments, and human liver type PFK cDNA. Only the HuPFK-2.5Kb DNA probe was shown to be useful in this study. After examining 30 human subjects, except a case with hypertriglyceridemia, no polymorphism was observed using the restriction enzymes BamHI, EcoRI, HaeIII, HindIII, KpnI, MspI and PstI. In addition, five established malignant cell lines were also subject to RFLP analysis and, except the TSGH cell line, all revealed the same pattern as that found in normal human subjects. These results suggested that the 5' region sequence of human PFK gene is rather conserved during evolution and that can be explained as retaining the catalysic site of PFK. |
