中文摘要 | 本研究乃以Cephalosporium acremonium菌株進行種菌培養,再經醱酵培養後,由醱酵槽取出之醱酵液進行頭孢菌素C的濃縮及純化。所得醱酵液利用本文所提案的冷凍濃縮法,由4,000毫升的醱酵液中,祇需取出1,500毫升,即可濃縮得原含量的80%。將濃縮液或原來醱酵液以選用的AmberliteXAD-4管住進行純化,所得頭孢菌素C的純度達96.36%。判斷本研究的產品,具有較高之純度。於不同樹脂吸附容量下測其回收率,得知在82%,及10%容量下可分別回收87%及95%之頭孢菌素C。 |
英文摘要 | This research involved the cultivation and fermentation of Ceρhalosporium acremonium. Cephalosporin C was purified and concentrated from the broth. 80% of the cephalosporin C was recovered by the cooling method, taking only 1,500 ml from 4,000 ml of broth. The broth which has been concentrated by the cooling method was further purified by a column of amberlite XAD-4. The purity was 96.36% evaluated from the relative area from HPLC. Itwas found, from inspecting different capacities of resin, that 87% or 95% can be recovered for the capacity of 82% or 10% respectively. |
