中文摘要 | 為加速繁殖幼苗以供企業化生產珍貴中藥材-臺灣金線蓮(Anoectochilus formosanus, Hayata),本研究先行選定不同培養基配方,結果發現1/2 MS之效果最佳。再以1/2 MS為基本成分,挑選最佳之生長調節劑組合。試驗結果顯示,2, 4-D利於芽球體類似物之生成;加入低濃度NAA(0.6ppm)利於根系之發育;BA(約3~5ppm)利於分支之發生及莖部之粗壯。又利用25/20℃變溫配合14/10小時之明/暗期,可促進金線蓮之抽苔開花,但加噴GA₃ 10 ppm則抑制之。開花後需加人工授粉方能結實。種子無菌播種繁殖所試之五種配方中,以1/2 MS加2 ppm NAA及1 ppm kinetin之培養效果最好。 |
英文摘要 | The purpose of this study was to select culture medium to accelerate the in vitro propagation of Anoectochilus formosanus Hayata, one of the most precious Chinese herb drugs. One-half strength of MS medium was confirmed as having the most suitable basal salt composition for the culture of stem segments judged by its effect on seedling growth. Various combinations of plant growth regulators were then tested with 1/2 strength MS basal salts in order to understand the influences on plant growth and development. Experimental results indicated that 2,4-D favored the production of protocorm-like bodies. Low concentration of NAA supplemented to the medium stimulated the development of root system. BA had the effects of producing branches are increasing diameter of the cultured stem. Photoperiodic experiment indicated that no symptom of flower-bud initiation was observed under light/dark period of 20/4 hr. Growing plants at day/night temperature of 25/20℃ and light/dark period of 14/10 hr stimulated bolting and blooming. Spraying GA₃ at a concentration of 10 ppm showed inhibitory effect on flower bud formation. In vitro propagation of seedlings from seeds could be easily done. However, it is also closely associated with the composition of culture medium. Among the five combinations of media tested, 1/2 strength MS basal salts supplemented with 2 ppm NAA and 1 ppm kinetin showed the best result. |
